目的 :观察补肾益精方药对老年大鼠细胞染色体端粒DNA重复序列长度的影响 ,探讨其延缓衰老的机理。方法 :以经方左归饮及自拟方益肾宝为补肾益精方药给予老年大鼠。以本实验室建立的PCR法测定老年大鼠肝、肾、睾丸及白细胞染色体端粒DNA重复序列的相对长度。用紫外分光光度计测端粒DNA扩增产物 ,以光密度值表示端粒DNA重复序列相对长度。结果 :左归饮和益肾宝两组大鼠除睾丸组织外 ,肝、肾和白细胞端粒DNA扩增产物光密度值均大于老年对照组 ,统计学处理具有显著性差异。结论 :左归饮和益肾宝补肾益精方药可使体细胞染色体端粒DNA重复序列减缓缩短 ,这可能就是补肾益精方药延缓机体衰老的作用机制之一。
Objective : To observe the effect of Bushen Yijing Prescription on the length of chromosomal telomere DNA repetitive sequences in aged rats, and to explore its anti-aging mechanism. Method : The aged rats were treated with prescriptions of Zuogui Decoction and Fangyishenbao for Bushenyijing prescription. The relative lengths of telomere DNA repeats in liver, kidney, testis and leukocyte chromosomes of aged rats were determined by PCR established in our laboratory. The telomere DNA amplification product was measured with an ultraviolet spectrophotometer, and the relative length of the telomere DNA repeat sequence was expressed as the optical density value. Results : In addition to testicular tissue, the optical density of telomere DNA amplification products in liver, kidney and leukocyte were higher in the two groups than those in the old control group. There was significant difference in statistical treatment between Zuoguiyin and Yishenbao. Conclusion : Zuoguiyin and Yishenbaoshenjingyijing can slow down and shorten the telomere DNA repeat sequence of somatic cells, which may be one of the mechanisms of Bushen Yijing prescription delaying body aging.
