目的探讨塞来昔布对子宫内膜癌细胞HEC-1-B的作用及对COX-2 mRNA表达的影响。方法采用细胞培养、MTT试验研究不同浓度的塞来昔布对人类子宫内膜癌细胞HEC-1-B生长的抑制作用,通过RT-PCR法检测塞来昔布作用后COX-2 mRNA表达的变化。结果塞来昔布能够有效地抑制HEC-1-B细胞的生长,并呈一定的剂量和时间依赖关系,同时能够抑制其COX-2 mRNA表达,20μmol/L和50μmol/L塞来昔布对COX-2mRNA表达的抑制率分别为25.92%和50.81%。结论塞来昔布可能是通过降低COX-2 mRNA的表达而抑制子宫内膜癌细胞的生长。
Objective To investigate the effect of celecoxib on endometrial carcinoma cell line HEC-1-B and its effect on the expression of COX-2 mRNA. Methods Cell culture and MTT assay were used to study the inhibitory effect of celecoxib on the growth of human endometrial carcinoma cell line HEC-1-B. The expression of COX-2 mRNA was detected by RT-PCR after treated with celecoxib Variety. Results Celecoxib could effectively inhibit the growth of HEC-1-B cells in a dose-dependent and time-dependent manner while inhibiting COX-2 mRNA expression, 20 μmol / L and 50 μmol / L of celecoxib The inhibition rates of COX-2 mRNA expression were 25.92% and 50.81% respectively. Conclusion Celecoxib may inhibit the growth of endometrial cancer cells by decreasing the expression of COX-2 mRNA.
