研究单纯疱疹病毒侵入介质(herpesvirus entry mediator,HVEM)在人CD4~+CD25~+调节性T细胞(Treg)上的表达及对Treg功能的影响。分离人外周血单个核细胞,磁珠纯化获得T细胞及其不同亚群,流式细胞术检测Treg上HVEM的表达。Treg按不同比例分别与去除Treg的CD3~+T细胞、CD4~+CD25~-T细胞或CD8~+T细胞混合培养,加入抗HVEM抗体或同型对照抗体,并用CD3抗体单独或联合CD28抗体刺激T细胞,MTT法检测T细胞增殖。结果显示,Treg上HVEM表达量为90%~95%(n=15)。抗HVEM抗体组与对照组相比,降低了Treg对各类T细胞增殖的抑制作用(P<0.05),证明Treg表达的HVEM介导了其发挥抑制功能。
To study the expression of HVEM in human CD4 ~ + CD25 ~ + regulatory T cells (Tregs) and its effect on the function of Tregs. Peripheral blood mononuclear cells were isolated and T cells and their subpopulations were purified by magnetic beads. The expression of HVEM on Tregs was detected by flow cytometry. Tregs were mixed with Treg-depleted CD3 + T cells, CD4 + CD25 + T cells or CD8 + T cells in different ratios and anti-HVEM antibody or isotype control antibody was added and stimulated with CD3 antibody alone or in combination with CD28 antibody T cells, MTT assay T cell proliferation. The results showed that the expression of HVEM on Treg was 90% -95% (n = 15). Compared with the control group, anti-HVEM antibody group reduced the inhibitory effect of Treg on the proliferation of various T cells (P <0.05), demonstrating that Treg-expressing HVEM mediates its inhibitory function.
