目的:通过观察低分子量肝素(LMWH)对小鼠凝血时间及大鼠静脉血栓形成的影响,对其主要药效学进行了研究,并与国外同类产品Clivarin及普通肝素进行了比较。方法:玻片法体外测定小鼠全血凝血时间及大鼠体内静脉血栓形成法。结果:LMWHsc,450,150及50u/kg可明显延长小鼠全血凝血时间;sc,100,30u/kg可明显降低大鼠静脉血栓重量。与相同剂量Clivarin比较,二者药效相当;与普通肝素比较,LMWH有效剂量明显低于普通肝素。结论:LMWH具有生物利用度高、抗栓作用强及对凝血系统影响小的特性。
OBJECTIVE: To observe the effect of low molecular weight heparin (LMWH) on clotting time and venous thrombosis in rats. The main pharmacodynamics of LMWH were compared with Clivarin and unfractionated heparin. Methods: The whole blood clotting time in mice was determined by slide method and venous thrombosis in rats. Results: LMWHsc, 450,150 and 50u / kg could significantly prolong the whole blood clotting time of mice; sc, 100,30u / kg could significantly reduce the weight of venous thrombus in rats. Compared with the same dose of Clivarin, the efficacy of the two was comparable; compared with unfractionated heparin, the effective dose of LMWH was significantly lower than that of unfractionated heparin. Conclusion: LMWH has the characteristics of high bioavailability, strong antithrombotic effect and little influence on the coagulation system.
