目的探讨酰基缩肽1(ADEP1)对肾癌细胞凋亡及SHH信号通路相关蛋白Gli-1和Bcl-2表达的影响。方法 50μmol/L ADEP1处理肾癌ACHN细胞和769-P细胞48 h,采用光学显微镜观察细胞凋亡的细胞形态学和细胞核形态变化,MTT法检测细胞增殖,实时定量PCR(qRT-PCR)检测SHH信号通路SHH和Gli-1 mRNA的表达,Western blot法检测Gli-1和Bcl-2蛋白的表达。结果 ADEP1可诱导肾癌细胞发生凋亡,降低SHH和Gli-1 mRNA表达,以及Gli-1和Bcl-2蛋白的表达。结论ADEP1可通过抑制SHH信号通路相关蛋白表达,诱导肾癌细胞发生凋亡。
Objective To investigate the effect of acyl depsipeptide 1 (ADEP1) on the apoptosis of renal cell carcinoma and the expression of Gli-1 and Bcl-2 in SHH signaling pathway. Methods ACHN cells and 769-P cells were treated with 50 μmol / L ADEP1 for 48 h. Morphological and nuclear morphological changes were observed with light microscope. Cell proliferation was measured by MTT assay. SHR was detected by real-time quantitative PCR (qRT-PCR) The expression of Gli-1 and Bcl-2 proteins was detected by Western blot. Results ADEP1 induced cell apoptosis in renal carcinoma cells, decreased the expression of SHH and Gli-1 mRNA, and the expression of Gli-1 and Bcl-2 proteins. Conclusion ADEP1 can induce the apoptosis of renal carcinoma cells by inhibiting the expression of SHH signaling pathway related proteins.
