以抗轮纹病苹果砧木J35-8的茎尖为试材,通过筛选合适的培养基,研究了培养基中不同植物生长调节剂配比对芽苗诱导、增殖和生根的影响。结果表明:以MS为基本培养基,附加蔗糖35g/L、琼脂6.2g/L,适宜启动培养的植物生长调节剂配比为6-BA 1.00mg/L+NAA 0.05mg/L;适宜增殖培养的植物生长调节剂配比为6-BA 1.0mg/L+NAA 0.050mg/L,增殖系数可达到每4周8.50倍;以1/2MS为基本培养基,附加蔗糖15g/L、琼脂6g/L,适宜组培苗生根的植物生长调节剂配比为IAA 1.0mg/L+IBA 0.3mg/L,生根率达92.68%以上,每株苗平均生根9.35条,移栽成活率达100.00%。
The effects of different plant growth regulators on the induction, proliferation and rooting of sprouts were studied by selecting appropriate shoots from the shoot tip of apple rootstock J35-8. The results showed that with MS as the basic medium, 35g / L Sucrose and 6.2g / L agar were added, and the suitable ratio of plant growth regulators to 6-BA 1.00mg / L and NAA 0.05mg / L was appropriate. Of plant growth regulators is 6-BA 1.0mg / L + NAA 0.050mg / L, the multiplication coefficient can reach 8.50 times every 4 weeks; with 1 / 2MS as the basic medium, add sucrose 15g / L, agar 6g / L, and the ratio of plant growth regulators suitable for tissue culture was 1.0mg / L IAA and 0.3mg / L IBA. The rooting rate reached 92.68%. The average number of rooting per plantlets was 9.35 and the survival rate of transplanting was 100.00%.
