目的阐明抵抗素、脂联素及其受体在胰岛素抵抗发生中的作用。方法30只Wistar雄性大鼠随机分为对照组和高脂组。用逆转录聚合酶链反应和Southern blot方法分析抵抗素、脂联素和脂联素受体表达的改变。结果高脂组大鼠体重明显增加,空腹血糖、低密度脂蛋白胆固醇、总游离脂肪酸、胰岛素和HOMA胰岛素抵抗指数明显高于对照组,高密度脂蛋白胆固醇明显低于对照组,糖耐量和胰岛素耐量明显下降(P均<0.01)。高脂组脂肪组织抵抗素和脂联素的表达均明显下降(P<0.01);脂联素受体1的表达呈下降趋势,但与对照组相比无统计学意义(P>0.05)。结论抵抗素和脂联素表达的下降在高脂饮食导致的胰岛素抵抗的发生中起着重要作用,脂联素受体1水平的降低可能是脂联素敏感性降低的重要原因之一。
Objective To elucidate the role of resistin, adiponectin and their receptors in the development of insulin resistance. Methods Thirty Wistar male rats were randomly divided into control group and hyperlipidemic group. The changes of resistin, adiponectin and adiponectin receptor expression were analyzed by reverse transcription polymerase chain reaction and Southern blot. Results The body weight of rats in high-fat group increased significantly. The fasting blood glucose, low density lipoprotein cholesterol, total free fatty acid, insulin and HOMA insulin resistance index were significantly higher than those in the control group. The levels of high density lipoprotein cholesterol were significantly lower than those in the control group. The tolerance decreased significantly (P <0.01). The expressions of adiponectin and adiponectin in adipose tissue were significantly decreased in hyperlipidemic group (P <0.01). The expression of adiponectin receptor 1 showed a decreasing trend, but there was no significant difference compared with that in control group (P> 0.05). Conclusions The decrease of resistin and adiponectin expression plays an important role in the development of insulin resistance induced by high fat diet. The decrease of adiponectin receptor 1 may be one of the important reasons for the decrease of adiponectin sensitivity.
