用基因转染的方法,建立稳定表达Smad7基因的永生化及恶性化人支气管上皮细胞系,用MTT法检测Smad7基因过表达对细胞生长、增殖的影响及对TGF β1介导的生长抑制效应的影响.结果表明在永生化和恶性化人支气管上皮细胞系BEP2D和BERP35T2中各筛选到2个稳定表达SMAD7蛋白的克隆,命名为BS7 1、BS7 2(来源于BEP2D),RS7 1、RS7 2(来源于BERP35T2).这些细胞系细胞的生长、增殖能力均强于转染空载体的对照细胞系,同时TGF β1对这些细胞系的生长抑制能力明显减弱.提示Smad7基因通过降低细胞对TGF β的应答来促进细胞的生长、增殖能力.
To establish an immortalized and malignant human bronchial epithelial cell line stably expressing Smad7 gene by gene transfection method and detect the effect of Smad7 overexpression on cell growth and proliferation and the effect of TGF β1-mediated growth inhibition by MTT assay The results showed that two clones stably expressing the SMAD7 protein were screened in the immortalized and malignant human bronchial epithelial cell lines BEP2D and BERP35T2 and named as BS7 1, BS7 2 (derived from BEP2D), RS7 1, RS7 2 Derived from BERP35T2) .Cell growth and proliferation of these cell lines are stronger than control cells transfected with empty vector, while TGFβ1 significantly inhibited the growth inhibition of these cell lines, suggesting that Smad7 gene by reducing the cells of TGFβ Response to promote cell growth and proliferation.
