长非编码RNA(long non-coding RNA,lnc RNA)是真核生物中广泛存在的一类转录本超过200 nt不编码蛋白质的RNA,参与干细胞维持、胚胎发育、细胞分化、细胞生长、细胞凋亡、代谢、信号传导以及免疫应答等生理或病理过程的调控。本研究结合植物基因组特点以及高通量RNA测序数据,采用生物信息学手段建立并优化了一套植物lnc RNA筛选和鉴定的方法。利用此方法在胡杨异形叶发生的138 845个RNA-Seq测序数据中,筛选并鉴定了2 448个参与胡杨异形叶发生的lnc RNA,并进一步对其功能进行预测和分析,为研究胡杨异形叶生长发育特性及最终揭示叶形发育的分子机制提供资料和理论帮助。
Long non-coding RNA (lnc RNA) is a type of transcript that is widely found in eukaryotes. Over 200 nt of non-coding RNAs are involved in stem cell maintenance, embryonic development, cell differentiation, cell growth, Death, metabolism, signal transduction and immune response and other physiological or pathological process regulation. In this study, a set of plant lnc RNA screening and identification methods were established and optimized by using bioinformatics methods combined with plant genome features and high-throughput RNA sequencing data. Using this method, we screened and identified 2 448 lnc RNAs involved in the development of alien leaf of 138 Populus euphratica in 138 845 RNA-Seq sequences, and further predicted its function. Growth and development characteristics and finally reveal the molecular mechanism of leaf development to provide information and theoretical help.
