目的探讨Bcl-2和PTEN在宫颈癌组织、宫颈上皮内瘤样病变(CIN)和正常宫颈组织中表达情况及意义。方法采用组织芯片技术和免疫组织化学S-P法检测50例宫颈癌、31例CIN和20例正常宫颈组织中PTEN和Bcl-2的表达情况。结果 Bcl-2和PTEN表达定位于细胞浆和细胞核,其在宫颈癌中阳性表达率分别为82%(41/50)和32%(16/50),在CIN中的表达率分别为77%(24/31)和52%(16/31),在正常宫颈组织中表达率分别为0(0/20)和80%(16/20);两者在宫颈癌的表达情况均与淋巴结转移、临床分期及组织学分化程度相关(P均<0.05);在宫颈癌组织中Bcl-2和PTEN表达呈负相关(P<0.05)。结论随着癌前病变的进展及宫颈癌临床分期升高、病理组织学分化程度降低和淋巴结转移,PTEN和Bcl-2表达增强,说明两者与宫颈癌的发生、发展和转移密切相关。
Objective To investigate the expression and significance of Bcl-2 and PTEN in cervical cancer, cervical intraepithelial neoplasia (CIN) and normal cervical tissue. Methods The expressions of PTEN and Bcl-2 in 50 cases of cervical cancer, 31 cases of CIN and 20 cases of normal cervical tissue were detected by tissue microarray and immunohistochemical S-P method. Results The positive expression rate of Bcl-2 and PTEN in the cytoplasm and nucleus were 82% (41/50) and 32% (16/50) in cervical cancer respectively, and the positive rates in CIN were 77% (24/31) and 52% (16/31) respectively. The expression rates in normal cervical tissues were 0 (0/20) and 80% (16/20), respectively. The expressions of both in cervical cancer and lymph node metastasis (P <0.05). There was a negative correlation between the expression of Bcl-2 and PTEN in cervical cancer (P <0.05). Conclusions With the progression of precancerous lesions and the clinical stage of cervical cancer, the histological differentiation and lymph node metastasis are decreased, the expressions of PTEN and Bcl-2 are enhanced, which indicates that both of them are closely related to the occurrence, development and metastasis of cervical cancer.
