目的 初步探讨甲状腺乳头状癌 (PTC)中 p5 3基因突变及其与 p2 1WAF1 /CIP1 蛋白定量表达的关系。 方法 采用 PCR- SSCP、DNA序列分析、免疫组织化学、图像分析等技术 ,检测 4 1例 PTC和 10例甲状腺滤泡性腺瘤标本组织中 p5 3基因突变情况和 p2 1蛋白染色状况。 结果 (1)伴 p5 3基因突变 ( 组 )和不伴突变 ( 组 ) PTC的原发肿瘤 (T)、淋巴结转移 (N)及远处转移 (M)差异无显著性 (P>0 .0 5 )。 (2 ) 、 和 之间 , 和 、 和 、 和 的平均积分光密度值 (D)差异具有显著性意义 (P<0 .0 1)。 结论 (1) PTC的演进和 p5 3基因突变与否无直接关系 ,p5 3蛋白的空间构象可能发挥关键作用。 (2 )在 PTC的发生、发展中存在依赖野生型 p5 3蛋白诱导P2 1WAF 1 /CIP1表达的途径
Objective To investigate the relationship between the mutation of p5 3 gene and the quantitative expression of p2 1WAF1 / CIP1 protein in papillary thyroid carcinoma (PTC). Methods PCR-SSCP, DNA sequence analysis, immunohistochemistry, image analysis and other techniques were used to detect the mutation of p5 3 gene and p21 protein in 41 specimens of PTC and 10 specimens of thyroid follicular adenoma.
