目的探讨低功率射频联合长春瑞滨长循环热敏脂质体对兔VX2肿瘤的抑瘤效果。方法应用超声引导下组织块包埋法建立新西兰大白兔股部肌肉内VX2肿瘤模型。待肿瘤生长至0.8~1.0 cm时,根据肿瘤大小按区组随机的方法将24只荷瘤兔分成4组。对照组(CON):静脉注射等量生理盐水;单纯射频组(RF):给予1 W 30 min射频治疗;长春瑞滨长循环热敏脂质体组(LCTSL):给予1.8 mg/kg长春瑞滨长循环热敏脂质体;长春瑞滨长循环热敏脂质体联合射频组(LCTSL+RF):射频作用(1 W 30 min)前给予1.8 mg/kg长春瑞滨长循环热敏脂质体。治疗分别在第1、4、7天,共3次。治疗前及治疗后超声连续观测肿瘤的大小,治疗结束后8 d,处死实验兔测量肿瘤重量,计算抑瘤率,标本进行病理检查。结果所有实验兔均能耐受治疗完成实验。LCTSL+RF组抑瘤率为87.2%,显著高于RF和LCTSL组,差异有统计学意义(P<0.05);单纯射频作用无明显抑瘤效果。病理检查显示单纯射频作用无明显肿瘤坏死,而长春瑞滨长循环热敏脂质体联合射频可见明显的肿瘤细胞损伤及坏死。结论低功率射频联合长春瑞滨热长循环热敏脂质体对兔VX2肿瘤有明显的抑瘤效果;射频消融联合长春瑞滨长循环热敏脂质体可能会增大消融范围,提高抗肿瘤疗效。
Objective To investigate the anti-tumor effect of low power radio frequency combined with vinorelbine long-circulating thermosensitive liposome on rabbit VX2 tumor. Methods VX2 tumor model was established in New Zealand White Rabbit femoral pits by ultrasound guided tissue embedding method. Tumor growth until 0.8 ~ 1.0 cm, according to tumor size by randomized block method 24 will be divided into four groups of tumor-bearing rabbits. Control group (CON): intravenous injection of equal amount of saline; RF group: 1 W 30 min radiofrequency treatment; vinorelbine long-circulating thermosensitive liposome group (LCTSL): 1.8 mg / kg vinorelbine (LCTSL + RF): 1.8 mg / kg vinorelbine long-circulating thermosensitive lipids were administered before radiofrequency (1 W for 30 min) Plastids. Treatment were in the first 1,4,7 days, a total of 3 times. Before treatment and after treatment, the size of the tumor was continuously observed by ultrasound. At 8 days after the treatment, the experimental rabbits were sacrificed to measure the tumor weight, and the tumor inhibition rate was calculated. The specimens were examined for pathology. Results All experimental rabbits were able to tolerate the treatment to complete the experiment. The inhibition rate of LCTSL + RF group was 87.2%, which was significantly higher than that of RF and LCTSL group (P <0.05). Pathological examination showed no significant tumor necrosis of the role of radio frequency alone, and vinorelbine long-circulating thermal liposomes combined with radiofrequency obvious tumor cell damage and necrosis. Conclusions Low-power radiofrequency combined with vinorelbine thermal long-circulating thermosensitive liposomes has obvious anti-tumor effect on rabbit VX2 tumors. Radiofrequency ablation combined with vinorelbine long-circulating thermosensitive liposomes may increase the scope of ablation and improve anti-tumor Efficacy.
