本实验用聚合酶链反应与随机引物标记法制备地高辛素标记的肾综合征出血热病毒核酸M、S片段cDNA探针。DNA-RNA斑点杂交结果表明，该探针具有肾综合征出血热病毒的特异性，可检出10pgHFRSV-RNA。实验结果表明：抗原阳性鼠体螨10只组、5只组，抗原阴性鼠体螨10只组，游离螨50只组、10只组，抗原阳性鼠肺50mg、100mg组，所抽提RNA与探针杂交结果呈阳性，而抗原阴性鼠体螨5只组抽提RNA的探针检测结果呈阴性。 In this study, cDNA probes of hemagglutinin-labeled M and S fragments of hemorrhagic fever with renal syndrome virus were prepared by polymerase chain reaction and random primer labeling. DNA-RNA dot blot results showed that the probe has the specificity of hemorrhagic fever with renal syndrome virus, can detect 10pgHFRSV-RNA. The results showed that there were 10 antigen-positive mouse mites in 10 groups, 5 groups, 10 antigen-negative mouse mites in 10 groups, 50 groups of free mites in 10 groups and 50 mg antigen- The result of hybridization was positive, while the probe of RNA extracted from 5 mice with antigen negative mite was negative.
