分别采用分子排阻色谱(SEC)法及其与多角度激光光散射法(SEC-MALLS)联用测定普鲁兰糖的相对分子量(Mw)。SEC法采用示差检测器(RI),SEC-MALLS法为MALLS与RI联用。采用t检验比较2种方法的分析结果,结果显示SEC-MALLS与SEC法测定结果一致。分别分析和评定这2种方法的不确定度,计算测定过程中的不确定度分量、合成不确定度和扩展不确定度。结果表明SEC-MALLS法稳定可靠,可替代SEC法。
The relative molecular weight (Mw) of pullulan was determined by size exclusion chromatography (SEC) and its combination with multi-angle laser light scattering (SEC-MALLS). SEC method using differential detector (RI), SEC-MALLS method for MALLS combined with RI. The t test was used to compare the results of the two methods. The results showed that the SEC-MALLS was consistent with the SEC method. Analyze and evaluate the uncertainty of these two methods separately, calculate the components of uncertainty in the determination process, synthesize the uncertainty and expand the uncertainty. The results show that the SEC-MALLS method is stable and reliable, which can replace the SEC method.
