目的:探讨肝癌组织中G蛋白偶联受体48(GPCR48)的表达与肝癌转移和预后的关系。方法:选择2019年1月至8月在广西国际壮医医院肝胆外科行手术切除(Rn 0切除)的10例肝癌患者,其中男性6例,女性4例,年龄37.0~64.0岁;选择2019年1月至12月在广西国际壮医医院肝胆外科行肝部分切除术的6例因外伤或结石行肝部分切除的患者,其中男性3例,女性3例,年龄16.0~63.0岁。收集肝癌患者的肝癌组织、癌旁组织及肝部分切除患者的正常肝组织,利用实时荧光定量PCR、蛋白印迹技术和免疫组化技术检测肝癌组织、癌旁组织和正常肝组织中GPCR48的mRNA和蛋白表达,比较分析肝癌组织、癌旁组织和正常肝组织中GPCR48表达的差异。选择2009年6月至2019年6月在南宁市第一人民医院肝胆外科行肝癌切除术的68例肝癌患者癌组织标本以及相关临床资料,其中男性56例,女性12例,年龄范围31.0~76.0岁,采用免疫组化技术检测肝癌组织中GPCR48的表达,根据肝癌组织中GPCR48的表达水平将肝癌患者分为GPCR48高表达组和GPCR48低表达组,分析GPCR48的表达水平与肝癌转移和预后的关系。n 结果:肝癌组织GPCR48 mRNA的表达水平高于癌旁组织[(0.894±0.086)比(0.583±0.095)]和正常肝组织[(0.894±0.086)比(0.485±0.111)],肝癌组织GPCR48蛋白的表达水平高于癌旁组织[(0.772±0.080)比(0.453±0.052)]和正常肝组织[(0.772±0.080)比(0.329±0.046)],差异均具有统计学意义(n P<0.05)。肝癌组织GPCR48蛋白阳性率为70.0%(7/10),癌旁组织为20.0%(2/10),两者差异具有统计学意义(n P<0.05)。GPCR48表达与肿瘤长径、TNM分期、血管侵犯、肿瘤包膜等相关(n P0.05)。68例肝癌患者中,GPCR48高表达组46例,GPCR48低表达组22例,GPCR48高表达组患者肿瘤累积复发率高于GPCR48低表达组(χn 2=15.577),累积生存率低于GPCR48低表达组(56.5%比27.3%,χn 2=6.309),差异均具有统计学意义(8.7%比36.4%,n P<0.05)。n 结论:GPCR48 mRNA和蛋白在肝癌组织中表达上调,与肝癌复发转移有关,影响肝癌患者的预后。“,”Objective:To study the correlations between GPCR48 expressions and metastasis and prognosis in hepatocellular carcinoma (HCC) patients.Methods:A total of 10 HCC patients underwent surgical resection (Rn 0 resection) at the Department of Hepatobiliary Surgery of Guangxi International Zhuang Medical Hospital from January to August 2019 were included. There are 6 males and 4 females, aging 37.0-64.0 years old. A total of six patients underwent partial hepatectomy suffering from trauma or stones at the Department of Hepatobiliary Surgery of Guangxi International Zhuang Medical Hospital from January to December 2019 were selected, including 3 males and 3 females, aged 16.0 to 63.0 years. The tumor and peritumoral tissues were collected, and the normal liver tissues were collected from trauma or calculus. The expressions of GPCR48 mRNA and protein of tumor, peritumoral, and normal liver tissues were detected using Real-time PCR, immunohistochemical, and Western blot, respectively. Comparative analysis of the differences of GPCR48 expression in tumor, peritumoral and normal liver tissues. Another 68 patients with HCC underwent hepatectomy from the Department of Hepatobiliary Surgery of the First People’s Hospital of Nanning from June 2009 to June 2019 were enrolled. There were 56 males and 12 females, with ages ranging from 31 to 76 years. Immunohistochemical was used to detect GPCR48 expressions in HCC tissues. The patients were divided into the GPCR48 high expression group and low expression group. The correlations between the expressions of GPCR48 and metastasis and prognosis of HCC were analyzed.n Results:The mRNA expressions in HCC tissues were significantly higher than that in the peritumoral tissues and normal liver tissues [(0.894±0.086) vs (0.583±0.095), (0.894±0.086) vs (0.485±0.111), n P<0.05], and the protein expressions in HCC tissues were also significantly higher than that in the peritumoral tissues, and normal liver tissues [(0.772±0.080) vs (0.453±0.052), (0.772±0.080) vs (0.329±0.046),n P<0.05]. The positicity of GPCR48 in HCC tissue was 70.0% (7/10), and the positicity was 20.0% in the adjacent tissues (2/10). The differences between the two groups were statistically significant (n P<0.05). The expression of GPCR48 was positively correlated with tumor size, TNM type, vascular invasion and tumor encapsulation (56.5% vs 27.3%,n P<0.05). There were 46 patients in the GPCR48 high expression group and 22 patients in GPCR48 low expression group. The tumor recurrence rate in the GPCR48 high expression group was significantly higher than that in the GPCR48 low expression group (8.7% vs 36.4%, χn 2=15.577, n P<0.05). while the survival rate was significantly lower in the GPCR48 high expression group than that in GPCR48 low expression group (χn 2=6.309, n P<0.05).n Conclusions:Expressions of GPCR48 were upregulated in HCC tissues, which were related to the recurrence and metastasis of liver cancer, and affected the prognosis of liver cancer patients.
