以南方红豆杉种子为试验材料,利用L_(25)(5~3)正交试验,采用GA_3、6-BA和2,4-D激素,浓度梯度为0、0.25、0.5、0.75和1.0 mg/L,测定种子的萌发率、发芽指数、活力指数和平均发芽时间,研究激素处理下对种子萌发的影响,探讨促进南方红豆杉种子萌发的有效激素浓度配比。试验结果表明,组合A_1B_2C_2的萌发率最高,为85.33%;A_1B_1C_1的发芽指数(6.95)和活力指数(19.25)最高;A_4B_2C_5的平均发芽时间最短,为6.04 d。综合分析,GA_30.75 mg/L+6-BA 0.25 mg/L对提高萌发率、缩短发芽时间效果最好;除平均发芽时间外,其他各指标的极差值均表现为GA_3>6-BA>2,4-D,平均发芽时间的极差值排序为6-BA>GA_3>2,4-D。GA_3、6-BA对种子的萌发率分别有极显著影响(P<0.01)和显著影响(P<0.05),主效应为GA_3>6-BA>2,4-D;GA_3、6-BA对种子的发芽指数、活力指数和平均发芽时间均有极显著影响(P<0.01),主效应为GA_3>6-BA>2,4-D;平均发芽时间的主效应为6-BA>GA_3>2,4-D。由此可知,GA_3是影响南方红豆杉种子萌发率、发芽指数及活力指数的关键因素,6-BA是促进南方红豆杉种子提早萌发的关键因素,2,4-D作用不明显。
Taxus chinensis seeds were used as experimental materials, GA3,6-BA and 2,4-D hormones were used in orthogonal experiment of L25 (5-3) with concentration gradient of 0,0.25,0.5,0.75 and 1.0 mg / L, the seed germination rate, germination index, vigor index and average germination time were measured to study the effect of hormone treatment on seed germination, and to explore the effective hormone concentration ratio to promote the seed germination of Taxus chinensis. The results showed that the germination rate of the combination A_1B_2C_2 was the highest (85.33%). The highest germination index (6.95) and activity index (19.25) of A_1B_1C_1 were the highest. The average germination time of A_4B_2C_5 was the lowest, 6.04 days. The results showed that GA_30.75 mg / L + 6-BA 0.25 mg / L had the best effect on increasing the germination rate and shortening the germination time. The average values of GA_3> 6-BA > 2,4-D. The average value of germination time ranked 6-BA> GA_3> 2,4-D. The main effects of GA 3,6-BA on the seed germination rate were significant (P <0.01) and significant (P <0.05), respectively, and the main effects were GA 3> 6-BA> 2,4-D and GA 3,6-BA The main effect of seed germination index, vigor index and average germination time were GA3> 6-BA> 2,4-D. The main effect of average germination time was 6-BA> GA3> 2,4-D. Thus, GA_3 was the key factor affecting the seed germination rate, germination index and vigor index of Taxus chinensis var. Mairei. 6-BA was the key factor to promote the early germination of Taxus chinensis seed, but the effect of 2,4-D was not obvious.
