草丁膦作为转基因筛选剂具有高效、快速、简便等优点。为了提高筛选转基因大豆阳性植株的效率,以6个常用的转基因受体大豆品种为材料,在温室和田间,采用叶片涂抹和整株喷施两种方法,通过受体品种叶片颜色变化和致死浓度确定了转基因后代材料适宜筛选浓度,其中适宜涂抹浓度为100~150 mg·L-1,适宜喷施浓度为700~750mg·L-1。以转bar基因T1代大豆植株进行验证,筛选浓度筛选准确率分别达到61.3%和73.9%。本研究为抗草丁膦转基因大豆后代材料的筛选提供了重要依据。
Glufosinate as a GM screening agent with high efficiency, fast, simple and so on. In order to improve the efficiency of screening transgenic soybean-positive plants, six commonly used transgenic soybean varieties were used as materials in the greenhouse and fields. Two methods, leaf painting and whole plant spraying, were adopted to study the effects of leaf color change and lethal concentration The appropriate screening concentration of transgenic progeny was determined, and the suitable application concentration was 100-150 mg · L-1 and the suitable spraying concentration was 700-750 mg · L-1. The transgenic T1 plants of transgenic bar were validated, and the accuracy of screening concentration was 61.3% and 73.9% respectively. This study provides an important basis for the screening of glufosinate-resistant transgenic soybean progeny.
