目的:测定灵芝三萜平衡溶解度和表观油水分配系数,推测其体内吸收情况,并通过在体肠吸收进行验证。方法:将灵芝三萜划分为酸性三萜和中性三萜,采用可见分光光度法测定两者在不同pH溶液中的平衡溶解度及在正辛醇-水/缓冲液中的表观油水分配系数,选取几个成分考察其在不同肠段的消除率研究三萜的肠吸收特性。结果:37℃时酸性三萜和中性三萜的平衡溶解度均<100 mg.L-1,log P分别为1.83,1.52,两者口服肠吸收均可以,中性三萜相对较好。结论:灵芝酸性三萜和中性三萜水溶性均较差,溶出是其吸收的限速过程。
OBJECTIVE: To determine the equilibrium solubility and apparent oil-water partition coefficients of ganoderma lucidum triterpenes, and to estimate its absorption in vivo and to verify its absorption in vivo. Methods: The triterpenoids of Ganoderma lucidum were divided into acidic triterpenoids and neutral triterpenoids. The equilibrium solubility of the two compounds in different pH solutions and the apparent water-oil partition coefficient in n-octanol-water / buffer were determined by visible spectrophotometry , Select a few ingredients to study its intestinal elimination rate in different intestine intestinal absorption characteristics. Results: The equilibrium solubility of triterpenoids and neutral triterpenoids at 37 ℃ were all <100 mg.L-1, log P = 1.83,1.52, respectively. CONCLUSION: The water-soluble triterpenes and neutral triterpenes of Ganoderma lucidum are poor, and the dissolution rate is the rate-limiting process of their absorption.
