目的研究环氧合酶-2(COX-2)和基质金属蛋白酶-9(MMP-9)在宫颈癌组织中的表达及临床意义。方法采用免疫组化S-P法检测72例宫颈浸润癌(ICC)、16例癌旁正常宫颈上皮(NCE)组织中COX-2和MMP-9的表达,并分析COX-2和MMP-9表达与ICC病理分级、临床分期、间质浸润和淋巴结转移等临床病理特征的关系。结果ICC组织中COX-2和MMP-9的表达显著高于NCE组织,72例ICC中,COX-2阳性表达率为88.9%,MMP-9阳性表达率为94.4%;16例NCE组织中,COX-2阳性表达率为12.5%,MMP-9阳性表达率为43.8%,两组间差异均有统计学意义(P< 0.01)。COX-2表达与ICC的淋巴结转移和间质浸润深度呈正相关(r分别为0.296和0.257)。MMP-9的表达与ICC的临床分期、病理分级呈正相关(r分别为0.329和0.351)。COX-2和MMP-9在宫颈癌组织中的表达强度呈正相关(r=0.297),二者表达的一致率高达86.1%(62/72)。结论COX-2和MMP-9的高表达与宫颈癌的侵袭和发展有关,二者在宫颈癌的细胞增殖、浸润和转移中有协同作用,联合检测二者的表达对于进一步理解宫颈癌的生物学行为和判断预后有一定价值。
Objective To study the expression and clinical significance of cyclooxygenase-2 (COX-2) and matrix metalloproteinase-9 (MMP-9) in cervical cancer. Methods The expressions of COX-2 and MMP-9 in 72 cases of invasive cervical carcinoma (ICC) and 16 cases of normal cervical epithelium adjacent to cancer were detected by immunohistochemical SP method. The expressions of COX-2 and MMP-9 ICC pathological grade, clinical stage, interstitial infiltration and lymph node metastasis and other clinicopathological features. Results The expression of COX-2 and MMP-9 in ICC tissues was significantly higher than that in NCE tissues. The positive rate of COX-2 in ICC was 88.9% and the positive rate of MMP-9 in ICC was 94.4% The positive rate of COX-2 was 12.5% and the positive rate of MMP-9 was 43.8%. There was significant difference between the two groups (P <0.01). COX-2 expression was positively correlated with lymph node metastasis and depth of interstitial infiltration in ICC (r = 0.296 and 0.257, respectively). The expression of MMP-9 was positively correlated with the clinical stage and pathological grade of ICC (r = 0.329 and 0.351, respectively). The positive expression of COX-2 and MMP-9 in cervical cancer tissues was significant (r = 0.297), the concordance rate was 86.1% (62/72). Conclusions The overexpression of COX-2 and MMP-9 is related to the invasion and development of cervical cancer. Both of them have a synergistic effect on the proliferation, invasion and metastasis of cervical cancer. The combined detection of the expression of COX-2 and MMP- Learning behavior and prognosis have some value.
