【目的】观察脉复生对急性血瘀模型大鼠血液流变学及凝血功能的影响。【方法】选用60只SD大鼠随机分为空白对照组、模型对照组、阳性对照组(100 mg/kg阿司匹林)以及脉复生高、中、低剂量组(剂量分别为10、5、2.5 g·kg-1·d-1),连续灌胃给药1周。末次给药后,采用各大鼠注射肾上腺素加冰浴法复制急性血瘀模型,测定各组大鼠血液流变学、凝血功能、红细胞压积和沉降率、血小板和红细胞电泳时间、血小板聚集率;采用酶联免疫吸附法测定血浆P选择素、血栓调节蛋白含量。【结果】与空白对照组比较,模型对照组大鼠的凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、凝血酶时间(TT)均显著缩短(P<0.01),而血浆P选择素、血栓调节蛋白、纤维蛋白原(FIB)的含量均显著增加(P<0.05或P<0.01);与模型对照组比较,脉复生组可显著降低血液流变参数,延长凝血酶时间、凝血活酶时间和凝血酶原时间,降低纤维蛋白原含量、血浆P选择素、血栓调节蛋白(P<0.05或P<0.01);与空白对照组比较,模型对照组血小板和红细胞电泳时间显著缩短(P<0.05或P<0.01),脉复生组可延长血小板和红细胞的电泳时间(P<0.05或P<0.01)。【结论】脉复生通过介导血浆P选择素和血栓调节蛋白而改善急性血瘀大鼠的血液流变性和凝血功能。
【Objective】 To observe the effect of Maifujian on hemorheology and coagulation function in acute blood stasis model rats. 【Methods】 Sixty Sprague-Dawley rats were randomly divided into blank control group, model control group, positive control group (100 mg / kg aspirin) and high, medium and low doses of pulse recovery group (doses of 10, 5 and 2.5 g · Kg-1 · d-1), continuous gavage for 1 week. The blood rheology, coagulation function, hematocrit and sedimentation rate, platelet and erythrocyte electrophoresis time, platelet aggregation The plasma P-selectin and thrombomodulin levels were determined by enzyme-linked immunosorbent assay. 【Results】 Compared with the blank control group, the prothrombin time (PT), activated partial thromboplastin time (APTT) and thrombin time (TT) of model control group were significantly shortened (P <0.01) P-selectin, thrombomodulin, fibrinogen (FIB) were significantly increased (P <0.05 or P <0.01). Compared with the model control group, the pulse recovery group could significantly reduce the parameters of blood rheology, prolong the thrombin time , Thromboplastin time, prothrombin time, fibrinogen content, plasma P-selectin and thrombomodulin (P <0.05 or P <0.01). Compared with the blank control group, the platelet and erythrocyte electrophoresis time of the model control group was significant (P <0.05 or P <0.01). The pulse recovery group could extend the electrophoresis time of platelet and erythrocyte (P <0.05 or P <0.01). 【Conclusion】 PPR can improve the hemorrheology and coagulation function of acute blood stasis rats by mediating the plasma P-selectin and thrombomodulin.
