目的:观察养精胶囊对老年雄性大鼠精囊超微结构的影响,探讨其促进精囊分泌功能的作用机制。方法:老年SD雄性大鼠50只,18~20月龄。随机分为空白对照组、十一酸睾酮组(4 mg/kg)、养精胶囊高(1.26 g/kg)、中(0.63 g/kg)、低(0.315 g/kg)剂量组共5组,每组10只。分别灌胃30 d后,摘取精囊,将精囊液挤入试管称重,测定各组精囊液果糖浓度,双侧精囊分别放入甲醛固定液和戊二醛中行组织学观察。结果:大鼠精囊腺脏器系数(g/g×106)、精囊液重量(g)及果糖浓度(mg/ml)分别为:空白对照组1 164.5±212.6、0.83±0.30、4.35±0.31;十一酸睾酮组1 510.5±313.1、0.82±0.25、5.35±0.71;养精胶囊高剂量组1 484.3±262.7、1.14±0.18、5.30±0.45;养精胶囊中剂量组1 396.6±268.9、0.83±0.24、4.71±0.41;养精胶囊低剂量组1 475.0±305.2、0.74±0.28、4.50±0.23。养精胶囊高剂量组能显著促进精囊液的分泌(P<0.05),养精胶囊中、低剂量组有改善趋势。苏木素伊红染色后镜下观察,养精胶囊组相对于空白对照组,上皮细胞增生更为活跃,细胞层次增多,细胞胞质丰富透明,内含有较多分泌颗粒、脂肪滴及脂褐素,腺腔腔缘模糊,腔内嗜伊红分泌物,其中结构改变以高剂量组最为显著。养精胶囊组分泌颗粒的面数密度与体密度与空白组比较具有统计学差异(P<0.05)。结论:养精胶囊可以通过改善精囊腺主细胞的分泌功能,进而促进精囊腺的分泌。
OBJECTIVE: To observe the effect of Yangjing Capsule on the ultrastructure of seminal vesicles in aged male rats, and to explore the mechanism of promoting the secretory function of seminal vesicles. METHODS: Fifty SD male rats aged 18 to 20 months were used. Randomly divided into blank control group, testosterone undecanoate (4 mg/kg), nourishing capsule high (1.26 g/kg), medium (0.63 g/kg), low (0.315 g/kg) dose groups of 5 groups , 10 in each group. After 30 days of gavage, the seminal vesicles were removed and the seminal vesicles were squeezed into tubes to measure the concentration of fructose in the seminal vesicles. The bilateral seminal vesicles were placed in formalin and glutaraldehyde for histological observation. Results: The visceral gland organ coefficient (g/g×106), seminal vesicle fluid weight (g) and fructose concentration (mg/ml) were: 164.5±212.6, 0.83±0.30, 4.35±0.31 in the blank control group; Testosterone undecanoate group 1 510.5±313.1, 0.82±0.25, 5.35±0.71; Yangjing capsule high dose group 1 484.3±262.7, 1.14±0.18, 5.30±0.45; Yangjing capsule middle dose group 1 396.6±268.9, 0.83± 0.24, 4.71±0.41; Yang Jing capsule low dose group 1 475.0±305.2, 0.74±0.28, 4.50±0.23. The high-dose group of Yangjing Capsule could significantly promote the secretion of seminal vesicle fluid (P<0.05), and the middle and low-dose groups of Yangjing Capsule had an improvement trend. After hematoxylin and eosin staining, the epithelial cell proliferation was more active in the Yangjing capsule group than in the control group. The number of cell layers increased, and the cytoplasm was rich and transparent, with more secretory granules, fat droplets, and lipofuscin. The lumen of the lumen of the gland is ambiguous, with intraluminal eosinophilic secretions, of which structural changes are most pronounced in the high-dose group. There was a statistically significant difference in surface density and body density of secretory granules in the Yang Jing capsule group compared with the blank group (P<0.05). Conclusion: Yangjing Capsule can promote the secretion of seminal vesicles by improving the secretion of seminal vesicles.
