目的 建立测定麻痹性贝毒素的细胞毒性试验。方法 体外培养小鼠神经母细胞瘤细胞 ,用石房蛤毒素为标准品 ,利用其钠通道阻断剂的特性 ,以四甲基偶氮唑蓝标定活细胞量 ,建立麻痹性贝毒素剂量与吸光度之间的标准曲线。结果 箭毒 /藜芦碱对照组OD值平均为 0 385 0 ,石房蛤毒素在 0 2~ 0 6ng剂量范围内与吸光度值呈直线相关关系 ,得回归方程为y =0 3996 +0 5 0 3x(r =0 9880 ,P <0 0 2 )。 结论 在 0~ 0 6ng剂量范围内 ,石房蛤毒素剂量与所测的OD值呈现剂量反应关系 ,该细胞毒性试验方法可用于麻痹性贝毒素的检测
Objective To establish a cytotoxicity test for paralytic shellfish poisoning. Methods The neuroblastoma cells were cultured in vitro. Using saxitoxin as a standard, the quantity of viable cells was established by tetramethylzidine blue Absorbance between the standard curve. Results The average OD value of the control group was 0 385 0, and saxitoxin had a linear correlation with the absorbance value at the dose range of 0 2 ~ 0 6 ng. The regression equation was y = 0 3996 +0 5 0 3x (r = 0 9880, P <0 0 2). Conclusion The dosages of saxitoxin in dose range of 0 ~ 0.6ng have a dose-response relationship with the measured OD value.
