①目的通过制备癌细胞凝集因子的单克隆抗体(McAb),进一步研究该凝集因子的结构、功能、组织来源,并为临床建立检测试剂盒打下基础。②方法采用杂交瘤技术将免疫小鼠脾细胞与SP2/O细胞融合,经ELISA法结合癌细胞凝集抑制实验筛选出阳性克隆。③结果融合阳性率78.1%;筛选出的杂交瘤细胞染色体数目为96~106条,SP2/0骨髓瘤细胞为68条,小鼠脾细胞为40条,得到的McAb亚类分别是IgG1(E9)和IgG2α(E11).④结论获得了2株既能分泌抗体,又能阻断癌细胞凝集的杂交瘤细胞株(E9和E11)。
Objective To further study the structure, function and tissue source of agglutination factor by preparing monoclonal antibody (McAb) for agglutination of cancer cells, and lay the foundation for establishing a detection kit for clinical practice. Methods The spleen cells of immunized mice were fused with SP2 / O cells by hybridoma technique. Positive clones were screened by ELISA and agglutination inhibition test. Results The positive rate of fusion was 78.1%. The number of hybridoma cells screened was 96-106, the number of SP2 / 0 myeloma cells was 68 and the number of spleen cells was 40. The McAb subclasses were IgG1 (E9) and IgG2α (E11). ④ Conclusion Two hybridoma cell lines (E9 and E11) that can both secrete antibodies and block the agglutination of cancer cells were obtained.
