目的:建立毛细管电泳法同时测定白芷药材中欧前胡素和异欧前胡素含量的方法。方法:采用未涂层弹性融硅石英毛细管柱55 cm×75μm ID,有效长度47 cm;以20 mmol/L NaH_2PO_4+100 mmol/L SDS+80%甲酰胺溶液(pH 7.12)为运行缓冲液;分离电压15 kV;重力进样5 s(高度15 cm);检测波长254 nm。结果:以盐酸小檗碱为内标,欧前胡素、异欧前胡素的线性范围分别为8.0~40.0μg/mL(r=0.9987)、4.0~20.0μg/mL(r=0.9990):白芷中欧前胡素的平均回收率为100.1%,方法精密度(RSD)为2.83%(n=6);异欧前胡素平均回收率为102.4%,方法精密度(RSD)为3.07%(n=6)。结论:该法简单、快速、准确可靠,可用于白芷药材的质量控制研究。
Objective: To establish a capillary electrophoresis method for the simultaneous determination of imperatorin and isoimperatorin in Radix Paeoniae Rubra. Methods: The uncoated elastic silica-silica capillary column was 55 cm × 75 μm ID, the effective length was 47 cm, and 20 mmol/L NaH 2 PO 4 +100 mmol/L SDS + 80% formamide solution (pH 7.12) was used as running buffer. Separation voltage 15 kV; gravity injection 5 s (height 15 cm); detection wavelength 254 nm. RESULTS: Using berberine hydrochloride as internal standard, the linear ranges of imperatorin and isoimperatorin were 8.0 to 40.0 μg/mL (r=0.9987) and 4.0 to 20.0 μg/mL (r=0.9990), respectively: The average recovery rate of the Central European Peucedanum was 100.1%, the method precision (RSD) was 2.83% (n=6), the average recovery rate of isoimperatorin was 102.4%, and the method precision (RSD) was 3.07% ( n=6). Conclusion: This method is simple, rapid, accurate and reliable. It can be used for the quality control of Radix Paeoniae Alba.
