目的研究乳浆大戟抑制人宫颈癌细胞增殖、迁移、侵袭和诱导细胞凋亡的作用并初步探讨其作用机制。方法用不同稀释度的乳浆大戟提取液处理人宫颈癌Si Ha细胞,MTT实验观察细胞生长抑制现象,吖啶橙染色荧光显微镜观察凋亡细胞的核形态改变并测定凋亡指数,流式细胞术检测细胞凋亡率,Transwell法检测细胞迁移能力和侵袭能力,用分光光度法检测caspase-3和caspase-9的表达情况。结果乳浆大戟对人宫颈癌Si Ha细胞有增殖抑制作用,该作用有时间和浓度依赖性。乳浆大戟处理后的人宫颈癌Si Ha细胞,凋亡指数和凋亡率均显著上升,均有时间和浓度依赖性。Transwell法检测显示,乳浆大戟提取液处理后的Si Ha细胞,迁移能力和侵袭能力显著下降。分光光度法检测发现,乳浆大戟处理后的Si Ha细胞中,caspase-3和caspase-9均表达增强。结论乳浆大戟提取液具有抑制人宫颈癌Si Ha细胞增殖和诱导细胞凋亡的作用,其作用通路有caspase-3和caspase-9的参与。
Objective To study the inhibitory effect of Escherichia coli on the proliferation, migration, invasion and apoptosis of human cervical cancer cells and to explore its mechanism. Methods Si Ha cells were treated with extract of Euphorbia pulcherrima with different dilutions. The growth inhibition of cells was observed by MTT assay. The apoptotic index of apoptotic cells was observed by acridine orange staining fluorescence microscope. Flow cytometry Cell apoptosis rate was detected by cytometry. Transwell assay was used to detect cell migration and invasion ability. The expression of caspase-3 and caspase-9 was detected by spectrophotometry. Results Escherichia coli inhibited the proliferation of human cervical cancer cells Si Ha cells in a time-and dose-dependent manner. Si Ha cells treated with the extract of Escherichia coli had a significant increase in apoptosis index and apoptotic rate, both in a time and concentration-dependent manner. Transwell assay showed that Si Ha cells treated with extract of Euphorbia pulcherrima decreased significantly in migration ability and invasiveness. Spectrophotometric analysis showed that expression of caspase-3 and caspase-9 were enhanced in Si Ha cells treated with the extract of Euphorbia miltiorrhiza. Conclusion Escherichia coli extract can inhibit the proliferation and induce the apoptosis of Si Ha cells in human cervical cancer cells. Caspase-3 and caspase-9 are involved in this pathway.
