[目的]探讨莪蚕健胃方对胃癌前病变大鼠MMP-2蛋白及其mRNA对血管生成的影响。[方法]实验大鼠分为正常组5只,造模组55只。制作胃癌前病变模型24周并抽检5只确保成功后,将剩余模型大鼠随机分为模型组,维甲酸组和中药高、中、低剂量组,每组10只。给药16周后处死取材。进行胃组织病理检测,MVD计数,和MMP-2蛋白及mRNA的检测。[结果]与正常组比较,造模组的MVD和MMP-2蛋白及其mRNA均过度表达(P<0.05),并随异型增生级别的升高而增强。MMP-2蛋白与MVD正相关(r=0.560,P<0.05),MMP-2mRNA与MVD无相关性(r=0.297,P>0.05)。在MVD、MMP-2蛋白及其mRNA的表达方面:与模型组相比,大部分治疗组能抑制其过度表达(P<0.05),中药低剂量组(P>0.05)除外。与维甲酸组相比,中药高剂量组表达降低(P<0.05),中药中剂量组差异无统计学意义(P>0.05)。[结论]1血管生成在胃癌前病变阶段即出现,MMP-2蛋白、mRNA呈过度表达,均随异型增生程度加重而增强。2大鼠胃癌前病变组织中MMP-2仅在蛋白水平与MVD呈正相关,提示MMP-2主要在蛋白层面参与了血管生成。3莪蚕健胃方可抑制胃癌前病变的血管生成,可能是通过削弱MMP-2基因的转录及表达而发挥治疗作用的。其作用与剂量有关。
[Objective] To investigate the effect of curcumin on the angiogenesis of rat gastric precancerous lesion rat MMP-2 protein and its mRNA. [Method] The experimental rats were divided into normal group (n = 5) and model group (n = 55). After 24 weeks of gastric precancerous lesion model development and 5 random sampling tests, the remaining model rats were randomly divided into model group, retinoic acid group and Chinese medicine high, medium and low dose group, 10 rats in each group. 16 weeks after the administration was sacrificed. Gastric histopathology, MVD count, and MMP-2 protein and mRNA were detected. [Results] Compared with normal group, MVD and MMP-2 protein and mRNA of model group were over-expressed (P <0.05), and increased with the increase of dysplasia level. There was a positive correlation between MMP-2 protein and MVD (r = 0.560, P <0.05). There was no correlation between MMP-2 mRNA and MVD (r = 0.297, P> 0.05). In MVD, MMP-2 protein and mRNA expression: compared with the model group, most of the treatment group can inhibit the over-expression (P <0.05), except for the low-dose group (P> 0.05). Compared with the retinoic acid group, the expression of high-dose Chinese medicine group decreased (P <0.05), while there was no significant difference between the medium-dose traditional Chinese medicine group (P> 0.05). [Conclusion] 1 Angiogenesis occurs in the precancerous stage of gastric cancer. MMP-2 protein and mRNA are overexpressed, all of which are increased with the degree of dysplasia. 2 MMP-2 protein in gastric precancerous lesions was positively correlated with MVD, suggesting that MMP-2 is mainly involved in angiogenesis at the protein level. 3 curcumin stomach can inhibit gastric precancerous angiogenesis, may be through the weakened MMP-2 gene transcription and expression and play a therapeutic role.
