目的:研究核内不均一性核糖核蛋白A1基因(heterogeneous nuclear ribonucleoproteins A1,hnRNP A1)mRNA水平在消化系统肿瘤组织中的表达及其临床意义。方法:利用TCGA数据库全基因组mRNA测序资料分析hnRNPA1在消化道肿瘤与正常组织间表达的差异,分析其表达水平与临床病理资料相关性,并进行生存分析判断其预后价值。结果:(1)hnRNPA1在不同消化系统肿瘤组织中表达较正常组织均明显增高(P<0.01),其中结直肠、肝癌、胆管癌、胰腺癌更为明显。(2)hnRNPA1在肝癌组织中的表达与肿瘤分化程度(P<0.05)、TNM分期(P<0.05)和浸润深度(P<0.05)相关,与年龄、性别、肝癌危险因素无关联。hnRNPA1在胆管癌、胰腺癌组织中的表达与年龄、性别、TNM分期、浸润深度等不相关。(3)hnRNPA1在胰腺癌组织中的表达高低与总生存期有相关性(P<0.05),且为独立于TNM分期、分化程度而影响患者预后的危险因子;hnRNPA1在结肠、直肠、肝、胆管肿瘤组织中的表达高低与总生存期无相关性。结论:hnRNPA1 mRNA在不同消化系统肿瘤中表达都增高;其高表达是胰腺癌独立预后不良因素,可以作为判断预后的有效生物标志物。
Objective: To investigate the expression of heterogeneous nuclear ribonucleoproteins A1 (hnRNP A1) mRNA in digestive system and its clinical significance. Methods: The difference of hnRNPA1 expression between gastrointestinal tumor and normal tissue was analyzed by genome-wide mRNA sequencing data of TCGA database. The correlation between hnRNPA1 expression and clinicopathological data was analyzed. The prognostic value of hnRNPA1 was analyzed by survival analysis. Results: (1) The expression of hnRNPA1 in different digestive system tumor tissues was significantly higher than that in normal tissues (P <0.01). Colorectal, liver cancer, cholangiocarcinoma and pancreatic cancer were more obvious. (2) The expression of hnRNPA1 in hepatocellular carcinoma was correlated with tumor differentiation (P <0.05), TNM stage (P <0.05) and depth of invasion (P <0.05), but not with age, sex and liver cancer risk factors. The expression of hnRNPA1 in cholangiocarcinoma and pancreatic cancer tissues was not related to age, gender, TNM stage, depth of invasion and so on. (3) The expression level of hnRNPA1 in pancreatic cancer was correlated with overall survival (P <0.05), and was a risk factor for prognosis in patients with TNM staging and differentiation; hnRNPA1 expression in colon, rectum, liver, There was no correlation between the expression of bile duct tumor and the overall survival. CONCLUSION: The expression of hnRNPA1 mRNA is increased in different digestive system tumors. High expression of hnRNPA1 mRNA is an independent prognostic factor for pancreatic cancer, which may be an effective biomarker for prognosis.
