【目的】观察中药抗病毒1号、2号胶囊对单纯疱疹病毒(HSV)-2感染骨髓间充质干细胞(BMSCs)Toll样受体9(TLR9)和干扰素(IFN)表达的影响。【方法】培养BMSCs进行HSV-2病毒接种,分别设立抗病毒1号、2号胶囊组,阿昔洛韦组,病毒组与空白对照组,采用Real-Time RT-PCR方法检测TLR9 mRNA表达,酶联免疫吸附(ELISA)法检测其下游产物IFN-α、IFN-β的表达。【结果】抗病毒1号、2号胶囊组TLR9 mRNA表达升高,IFN-α、IFN-β表达较病毒组显著升高(P﹤0.01),且抗病毒2号组作用优于抗病毒1号组(P﹤0.01)。【结论】抗病毒1号、2号胶囊治疗生殖器疱疹等病毒性疾病作用的可能机制与其能诱导TLR9及其下游产物IFN-α、IFN-β表达升高有关。
【Objective】 To observe the effect of Chinese traditional medicine anti-virus No.1 and No.2 capsules on the expression of TLR9 and IFN in HSV-2 infected bone marrow mesenchymal stem cells (BMSCs). 【Methods】 BMSCs were cultured and inoculated with HSV-2 virus. The anti-virus 1, 2 capsules, aciclovir group, virus group and blank control group were established. Real-time RT-PCR was used to detect TLR9 mRNA expression, The downstream products of IFN-α and IFN-β were detected by enzyme linked immunosorbent assay (ELISA). 【Results】 The expression of TLR9 mRNA in anti-virus No.1 and No.2 capsule groups was significantly higher than that in the control group (P <0.01), and the anti-virus 2 group was better than the anti-virus 1 Group (P <0.01). 【Conclusion】 The possible mechanism of anti-virus No. 1 and No. 2 capsules on the treatment of viral diseases such as genital herpes is related to its ability to induce the expression of TLR9 and its downstream products IFN-α and IFN-β.
