目的探讨肽基脯氨酰顺反异构酶1(Pin1)蛋白在宫颈鳞癌(CSCC)组织中的表达及与上皮间质转化相关蛋白E-钙粘素(E-cadherin)、N-钙粘素(N-cadherin)、β-连接蛋白(β-catenin)和波形蛋白(Vimentin)表达关系及意义。方法采用免疫组织化学SP法检测89例宫颈鳞癌组织及相应癌旁组织中Pin1、E-cadherin、Ncadherin、β-catenin和Vimentin蛋白表达。结果宫颈鳞癌组织中Pin1蛋白的阳性表达率(73.0%)明显高于正常宫颈组织(27.0%),且随着宫颈病变的进展,Pin1的表达逐渐增加,尤其在低分化鳞癌及伴有淋巴结转移的宫颈鳞癌中Pin1上调更加明显,差异有统计学意义(P<0.05);β-catenin、E-cadherin、N-cadherin和Vimentin蛋白在宫颈鳞癌中阳性表达率分别为37.1%、20.2%、66.3%和75.3%,随着宫颈病变的进展,上皮表面标志物E-cadherin和β-catenin表达明显下调(P<0.05),而间质细胞表型N-cadherin和Vimentin蛋白表达明显上调(P<0.05)。在宫颈鳞癌中Pin1与E-cadherin和β-catenin蛋白的表达呈负相关(r=-0.450,P<0.001;r=-0.367,P=0.003),而与N-cadherin和Vimentin蛋白的表达呈正相关(r=0.491,P=0.001;r=0.299,P=0.017)。结论宫颈鳞癌组织中Pin1蛋白表达上调可能是影响肿瘤细胞上皮间质转化相关蛋白E-cadherin、N-cadherin、β-catenin和Vimentin在细胞表达的重要原因之一,其可能反映宫颈鳞癌的发病机制。
Objective To investigate the expression of Pin1protein cis-1 (Pin1) protein in cervical squamous cell carcinoma (CSCC) and its relationship with the expression of E-cadherin, N-calcium (N-cadherin, β-catenin and vimentin expression and its significance. Methods The expressions of Pin1, E-cadherin, Ncadherin, β-catenin and Vimentin in 89 cases of cervical squamous cell carcinoma and corresponding paracancerous tissues were detected by immunohistochemical SP method. Results The positive rate of Pin1 protein in cervical squamous cell carcinoma was significantly higher than that in normal cervical tissue (73.0%, 27.0%). The expression of Pin1 increased with the progression of cervical lesions, especially in poorly differentiated squamous cell carcinoma and its associated The positive rate of Pin1 in cervical squamous cell carcinoma with lymph node metastasis was significantly higher than that in cervical squamous cell carcinoma with lymph node metastasis (P <0.05). The positive rates of β-catenin, E-cadherin, N-cadherin and Vimentin in cervical squamous cell carcinoma were 37.1% 20.2%, 66.3% and 75.3% respectively. With the progression of cervical lesions, the expressions of E-cadherin and β-catenin in epithelial surface were significantly decreased (P <0.05), while the expressions of N-cadherin and Vimentin in stromal cells were significantly (P <0.05). In Cervical squamous cell carcinoma, the expression of Pin1 was negatively correlated with the expression of E-cadherin and β-catenin (r = -0.450, P <0.001; r = -0.367, P = 0.003) (R = 0.491, P = 0.001; r = 0.299, P = 0.017). Conclusion The up-regulation of Pin1 protein in cervical squamous cell carcinoma may be one of the important factors affecting the expression of E-cadherin, N-cadherin, β-catenin and Vimentin in epithelial cells of cervical squamous cell carcinoma, Pathogenesis.
