目的:观察消痰散结方对MKN-45原位移植瘤裸鼠胃癌组织中成纤维细胞活化蛋白(fibroblast activation protein,FAP)表达的影响,探讨消痰散结方防止胃癌复发转移的作用机制。方法:建立人胃癌MKN-45裸鼠原位移植瘤模型,将40只模型鼠随机分为模型组、生理盐水组、中药组及化疗组,每组10只。观察各组荷瘤鼠肿瘤生长情况及抑瘤率;观察各组荷瘤鼠胃癌组织FAP蛋白及mRNA表达。结果:消痰散结方可明显抑制裸鼠人胃癌MKN-45原位移植瘤的生长,下调胃癌组织中FAP蛋白及mRNA的表达。结论:消痰散结方可以下调FAP蛋白及mRNA的表达,可能是其抗胃癌复发转移的机制之一。
Objective: To observe the effect of Xiaotan Sanjie Decoction on the expression of fibroblast activation protein (FAP) in gastric cancer of orthotopic transplanted MKN-45 in nude mice and to explore the mechanism of Xiaotan Sanjie Decoction preventing gastric cancer recurrence and metastasis . Methods: An in situ xenograft model of human gastric cancer MKN-45 in nude mice was established. Forty mice were randomly divided into model group, saline group, TCM group and chemotherapy group, with 10 in each group. The tumor growth and tumor inhibition rate in each group were observed. The expression of FAP protein and mRNA in gastric cancer tissues of each group were observed. Result: Xiaotan Sanjie can obviously inhibit the growth of orthotopic transplanted human gastric cancer MKN-45 in nude mice and down-regulate the expression of FAP protein and mRNA in gastric cancer. Conclusion: Xiaotansanjie Decoction can down-regulate the expression of FAP protein and mRNA, which may be one of the mechanisms of its anti-gastric cancer recurrence and metastasis.
