目的探讨Polo样蛋白激酶4(PLK4)表达与子宫内膜异位症(EMS)的相关性。方法 2013年5月至2014年1月在浙江大学医学院附属妇产科医院,采用免疫组织化学、实时荧光定量PCR技术(Real-time q PCR)以及蛋白免疫印迹的方法对37例异位子宫内膜(异位组)标本及32例正常子宫内膜(正常组)标本中PLK4的表达情况进行检测。结果免疫组织化学提示PLK4在异位组中的表达明显高于正常组,且差异有统计学意义(P<0.05)。PLK4 m RNA在异位组及正常组中均有表达,但异位组中的相对表达量(0.01190±0.00574)明显高于正常组(0.00227±0.00120),差异有统计学意义(P<0.01)。蛋白免疫印迹提示,PLK4蛋白在正常组及异位组内膜组织中均有表达,但异位组PLK4蛋白的表达显著高于正常组(P<0.01)。结论 PLK4在异位内膜组织中的高表达可能导致异位内膜加速生长,可能与EMS在临床上所表现出的恶性生物学行为有一定的关联。
Objective To investigate the relationship between Polo-like protein kinase 4 (PLK4) expression and endometriosis (EMS). Methods From May 2013 to January 2014, 37 cases of ectopic uterus were diagnosed by the method of immunohistochemistry, real-time q-PCR and Western blotting in the Affiliated Obstetrics and Gynecology Hospital of Zhejiang University School of Medicine. Endometrium (ectopic group) specimens and 32 cases of normal endometrium (normal group) specimens PLK4 expression was detected. Results Immunohistochemistry showed that the expression of PLK4 in ectopic group was significantly higher than that in normal group, and the difference was statistically significant (P <0.05). The expression of PLK4 mRNA in ectopic group and normal group was significantly higher than that in normal group (0.01190 ± 0.00574) (0.00227 ± 0.00120), the difference was statistically significant (P <0.01) . Western blotting indicated that PLK4 protein was expressed in normal and ectopic endometrium, but the expression of PLK4 protein in ectopic group was significantly higher than that in normal group (P <0.01). Conclusion The high expression of PLK4 in ectopic endometrium may result in the accelerated growth of ectopic endometrium, which may be related to the malignant biological behavior of EMS in clinic.
