目的:研究一水草酸钙(COM)晶体对肾小管上皮细胞的毒性作用。方法:以永生化的肾小管上皮HK-2细胞作为研究对象,用不同浓度的COM处理细胞,再以DAPI染色并细胞计数研究COM对HK-2细胞增殖的作用;检测细胞培养上清中的LDH含量,说明COM对HK-2的毒性;应用MTS法绘制HK-2细胞增殖曲线;使用流式细胞术检测COM对HK-2细胞的凋亡和坏死的作用。结果:DAPI染色后并细胞计数后HK-2细胞增殖能力经0.1mmol/L的COM处理后便受到抑制,并与对照组比较,差异有显著统计学意义,并随COM浓度的升高,抑制作用更加显著。1mmol/L的COM可使HK-2上清中的LDH显著升高,并在5mmol/L更加明显。1mmol/L的COM处理HK-2细胞后,细胞增殖曲线显著低于对照组,5mmol/L的COM在处理HK-2细胞的第6天几乎全部死亡。0.1mmol/L的COM可使HK-2细胞凋亡显著增加,1mmol/L的COM可使HK-2细胞坏死显著增加,并随浓度的升高而增加。结论:0.1mmol/L的COM刺激HK-2细胞即可使细胞出现生长抑制,浓度达到1mmol/L后,则可出现细胞死亡。
Objective: To study the toxic effects of COM crystals on renal tubular epithelial cells. Methods: Immortalized renal tubular epithelial HK-2 cells were treated with different concentrations of COM, then DAPI staining and cell counting were used to study the effect of COM on the proliferation of HK-2 cells. LDH content, indicating the toxicity of COM to HK-2; using MTS method to plot HK-2 cell proliferation curve; using flow cytometry to detect the effect of COM on HK-2 cells apoptosis and necrosis. RESULTS: After DAPI staining and cell counting, the proliferation of HK-2 cells was inhibited by 0.1 mmol / L COM treatment, and the difference was statistically significant compared with the control group, and with the increase of COM concentration, inhibition The effect is more significant. 1mmol / L of COM can significantly increase the LDH in HK-2 supernatant, and more obvious in 5mmol / L. After treated with 1 mmol / L COM for HK-2 cells, the cell proliferation curve was significantly lower than that of the control group. 5 mmol / L COM almost completely died on the 6th day after HK-2 cells were treated. 0.1 mmol / L COM could significantly increase the apoptosis of HK-2 cells, and 1 mmol / L of COM could significantly increase the necrosis of HK-2 cells and increase with the increase of concentration. CONCLUSIONS: 0.1 mmol / L COM stimulates HK-2 cells to inhibit the growth of cells. When the concentration reaches 1 mmol / L, cell death may occur.
