本研究以9份核桃属种质资源为材料,利用设计引物对其ITS区段进行PCR扩增和测序。利用Dnasp 5.0和MEGA 6.0软件分析ITS序列特征,寻找差异位点,并构建系统发育树。结果显示,9份材料的ITS区段在644~647 bp之间,存在48个变异位点,转换和颠换的比值为1.75。系统发育树结果表明,野核桃和核桃楸亲缘关系最近,最先聚在一起,再与麻核桃聚为核桃楸组,与传统分类学一致。本研究为发掘和利用核桃属种质资源奠定了理论基础。
In this study, 9 Walnut germplasm resources were used as materials, and their ITS regions were amplified by PCR and sequenced. Dnasp 5.0 and MEGA 6.0 software were used to analyze ITS sequence characteristics, find differences and construct phylogenetic tree. The results showed that 9 ITS segments ranged from 644 bp to 647 bp in length with 48 variation loci, and the ratio of transformation to transversion was 1.75. The results of phylogenetic tree showed that the relationship between wild walnut and Juglans manshuriensis had the closest relationship, the first clustered together, and then with the walnut walnut polygonatum group, consistent with the traditional taxonomy. This study laid the theoretical foundation for the exploration and utilization of walnut germplasm resources.
