水稻(Oryza sativa L.)作为粮食作物,在全世界粮食生产中具有极其重要的地位。稻瘟病、褐飞虱等病虫害的发生严重危害着水稻的安全生产,培育并利用水稻抗性品种能经济有效地预防水稻病虫害的发生,因此,培育抗性品种在水稻安全生产中尤为重要。本研究以具有抗稻瘟病基因Pi9的抗病品系75-1-127为抗病亲本,以携带有抗褐飞虱基因Bph18(t)的水稻材料C4064为抗虫亲本,以性状优良的恢复系测679作为轮回亲本,进行杂交、回交和自交,并辅以田间多代选择。在分离群体中,使用与Pi9紧密连锁的SCAR标记p B8跟踪目标基因Pi9,利用与Bph18(t)紧密连锁的标记KC16跟踪目标基因Bph18(t)。通过分子标记辅助选择、农艺性状评价和抗病抗虫鉴定,选育出聚合了Pi9和Bph18(t)基因、对稻瘟病和褐飞虱的抗性水平接近抗病亲本或抗虫亲本的恢复系。
Rice (Oryza sativa L.), as a food crop, plays an extremely important role in food production throughout the world. The occurrence of pests such as rice blast, brown planthopper and other pests seriously endangers the safe production of rice. To cultivate and utilize the resistant varieties of rice can effectively prevent the occurrence of rice pests and diseases. Therefore, it is very important to cultivate resistant varieties in the safe production of rice. In this study, the resistant strain 75-1-127 with the blast resistance gene Pi9 was used as the resistant parent and the rice material C4064 carrying the BPH gene Bph18 (t) was used as the insect-resistant parent. As a reincarnated parent, hybridization, backcrossing and selfing are complemented by multiple generations of field selection. In isolated population, the target gene Pi9 was traced using the SCAR marker pB8 closely linked to Pi9 and the target gene Bphl8 (t) was traced using the marker KC16 tightly linked to Bphl8 (t). Through molecular marker-assisted selection, agronomic traits evaluation and disease resistance and insect resistance identification, we screened out the restorer line that aggregated the genes Pi9 and Bph18 (t) and the resistance level to rice blast and brown planthopper was close to the resistant parent or insect-resistant parent.
