目的 探讨软肝散积方提取物 (AC - 0 1)预防和治疗肝纤维化的效果及部分作用机理。方法 采用CCl4化学性和猪血清免疫性诱导的大鼠肝纤维化模型 ,检测大鼠血清肝功能部分指标、肝组织羟脯氨酸 (HYP)含量、超氧化物歧化酶(SOD)活性 ,观察肝组织病理学并用SDS -PAGE、WesternBlotting测定I型胶原蛋白的表达。结果 不同剂量的AC - 0 1(1.34g/kg ,2 .6 8g/kg)可显著改善肝组织损伤和纤维化程度 ,同时能显著降低肝组织羟脯氨酸 (HYP)和I型胶原蛋白的含量 ,并且高剂量AC - 0 1(2 .6 8g/kg)的治疗作用更明显。结论 AC - 0 1有保护肝细胞 ,抗肝纤维化的作用。其作用机理与抗脂质过氧化损伤有关 ,对胶原代谢也有直接的影响
Objective To investigate the effect and some mechanism of action of Ruangan Sanjifang extract (AC - 01) in preventing and treating liver fibrosis. Methods The liver fibrosis model induced by CCl4 chemical and swine serum immunity was used to detect serum liver function, liver hydroxyproline (HYP) content and superoxide dismutase (SOD) activity. Liver histopathology and SDS-PAGE, Western Blotting were used to determine the expression of type I collagen. Results Different doses of AC-01 (1.34 g/kg, 2.8 g/kg) significantly improved liver injury and fibrosis, and significantly decreased liver hydroxyproline (HYP) and type I collagen. The effect of the high dose of AC-01 (2.68 g/kg) is more pronounced. Conclusion AC-01 protects hepatocytes against hepatic fibrosis. Its mechanism of action is related to anti-lipid peroxidative damage and has a direct effect on collagen metabolism.
