Plexin A1蛋白及mRNA在小鼠卵泡发育过程中的表达

目的:探讨Plexin A1在小鼠卵巢不同发育阶段的表达。方法:选择不同发育时期的C57BL/6j小鼠,采用免疫组织化学方法测定小鼠卵巢中Plexin A1蛋白水平的表达,实时定量聚合酶链反应(real-time PCR)方法检测Plexin A1mRNA水平的表达。结果:Plexin A1蛋白在小鼠卵巢各级卵泡的卵母细胞和颗粒细胞中均有表达。随着卵泡的不断发育,其在卵母细胞和颗粒细胞中的表达水平均逐渐增强。始基卵泡卵母细胞中的Plexin A1蛋白表达水平明显低于初级、次级和窦状卵泡的卵母细胞。始基卵泡颗粒细胞中未见明显Plexin A1蛋白表达,至初级卵泡阶段可见颗粒细胞开始有Plexin A1蛋白表达,其水平低于次级及窦状卵泡。闭锁卵泡中的Plexin A1呈强阳性表达,明显高于生长卵泡中的表达水平。Plexin A1mRNA在卵巢组织中的表达水平随周(日)龄呈先升高后降低的趋势,5d,7d,3周小鼠卵巢中Plexin A1 mRNA表达水平分别为3d小鼠的1.56、1.26、0.79倍(P<0.05),而2周小鼠卵巢中的Plexin A1mRNA表达水平与3d小鼠相当(P>0.05)。结论:Plexin A1在不同级别的小鼠卵泡中均有表达,且表达水平随卵泡的不断生长发育而增强,但闭锁卵泡中表达最强,提示Plexin A1在卵泡的生长发育及闭锁过程中可能发挥重要的调控作用。 Objective: To investigate the expression of Plexin A1 at different developmental stages of mouse ovary. Methods: C57BL / 6j mice were selected at different developmental stages. Immunohistochemistry was used to detect the expression of Plexin A1 protein in ovary of mice. The expression of Plexin A1 mRNA was detected by real-time PCR. Results: Plexin A1 protein was expressed in oocytes and granulosa cells of all ovarian follicles in mice. With the continuous development of follicles, their expression levels in oocytes and granulocytes are gradually increased. Plexin A1 protein expression in primordial follicular oocytes was significantly lower than that in primary, secondary and sinusoid follicles. No significant Plexin A1 protein was found in primordial follicular granulosa cells, and granulosa cells began to express plexin A1 protein in primary follicles, which was lower than that in secondary and sinusoid follicles. Plexin A1 in the atretic follicles was strongly positive, which was significantly higher than that in the growing follicles. The expression level of Plexin A1mRNA in ovary tissue firstly increased and then decreased with the rise of the week (day), and the expression levels of Plexin A1 mRNA in the ovary of mice on the 5th, 7th and the 3rd week were 1.56, 1.26 and 0.79 (P <0.05), while the expression level of Plexin A1 mRNA in the ovary of 2 weeks mice was similar to that of 3d mice (P> 0.05). Conclusion: Plexin A1 is expressed in different levels of mouse follicles, and its expression level increases with the continuous growth and development of follicles, but the strongest expression in atresia follicles, suggesting that Plexin A1 may play a role in follicular growth and atresia Important regulatory role.
