目的:观察豆腐果苷(helicid)对坐骨神经慢性压迫伤(CCI)后大鼠的镇痛作用及脊髓c-fos表达的影响.方法:32只Wistar雄性大鼠随机分为4组(n=8);分别为Sham组、CCI组、H1组和H2组。4组分别于CCI手术前1天、手术后第9天进行实验,测后肢的机械痛阈值(MWT)。H1组和H2组手术后10天开始给药,测定1、3、5、7、10天后肢MWT,第14天取大鼠脊髓腰膨大部位免疫组化染色检测Fos免疫阳性细胞。结果:豆腐果苷可明显提高CCI模型大鼠的疼痛阈值(P<0.05),抑制由疼痛所引起的脊髓背角c-fos表达增强的效应(P<0.05)。结论:口服豆腐果苷能剂量依赖性提高CCI大鼠疼痛阈值,降低机械痛觉过敏,抑制神经损伤后脊髓背角c-fos的表达。
Objective: To observe the analgesic effects of helicid on rats after chronic constriction injury (CCI) and the expression of c-fos in spinal cord.Methods: Thirty-two male Wistar rats were randomly divided into 4 groups (n = 8 ); Sham group, CCI group, H1 group and H2 group respectively. Four groups were given one day before CCI surgery and the other nine days after the operation. The mechanical pain threshold (MWT) of the hindlimb was measured. The H1 and H2 groups were administered 10 days after the operation, the hindlimb MWT was measured on the 1st, 3rd, 5th, 7th and 10th days. Fos immunoreactive cells were detected by immunohistochemical staining on the lumbar enlargement of the spinal cord on the 14th day. Results: Tofuoside significantly increased the pain threshold (P <0.05) and the effect of pain-induced enhancement of c-fos expression in spinal dorsal horn (P <0.05). Conclusion: Tocopherol can increase the pain threshold of CCI rats in a dose-dependent manner, reduce the mechanical hyperalgesia and inhibit the expression of c-fos in spinal dorsal horn after nerve injury.
