目的探讨扶正平喘汤防治过敏性哮喘的机制。方法将40只豚鼠随机分为正常对照组、哮喘模型组、地塞米松组、扶正平喘汤组,用鸡卵清蛋白雾化激发制成哮喘模型。地塞米松组与扶正平喘汤组分别于激发前3d给药,至激发后3d停药。用计数板计数外周血嗜酸性细胞(EOS)数量,观察小支气管形态学改变和肺组织中EOS浸润情况;生化法和放免法测定血清和肺泡灌洗液(BALF)中一氧化氮(NO)和内皮素-1(ET-1)的含量。结果扶正平喘汤可明显改善模型豚鼠小支气管损伤情况,减轻外周血和肺组织的EOS浸润程度,降低血清和BALF中NO的含量,降低BALF中ET-1的含量。结论扶正平喘汤可能通过减轻EOS浸润、调节机体NO和ET-1产生,对过敏性哮喘发挥预防和治疗作用。
Objective To explore the mechanism of Fuzheng Pingchuan decoction in preventing and treating allergic asthma. Methods Forty guinea pigs were randomly divided into normal control group, asthma model group, dexamethasone group, and Fuzheng Pingchuan decoction group. The asthma model was induced by chicken egg ovalin atomization. The dexamethasone group and the Fuzheng Pingchuan decoction group were administered 3 days before the challenge and were stopped 3 days after the challenge. Peripheral blood eosinophils (EOS) counts were counted using a counting plate to observe morphological changes in small bronchi and EOS infiltration in lung tissue. Biochemical and radioimmunoassay methods were used to determine nitric oxide (NO) in serum and bronchoalveolar lavage fluid (BALF). And endothelin-1 (ET-1) content. Results Fuzheng Pingchuan decoction can significantly improve the bronchiolar damage of model guinea pigs, reduce the degree of EOS infiltration in peripheral blood and lung tissue, decrease the content of NO in serum and BALF, and reduce the content of ET-1 in BALF. Conclusion Fuzheng Pingchuan Decoction may play a preventive and therapeutic role in allergic asthma by alleviating EOS infiltration and regulating the production of NO and ET-1 in the body.
