目的 检测抑癌基因TIP30蛋白在涎腺腺样囊性癌 (SaliveryAdenoidCysticCarcinoma ,SACC)中的表达。方法 选取我院 1 983年至今病理诊断为涎腺腺样囊性癌组织标本 2 0例 ,男性 9例 ,女性 1 1例 ;另选 8例正常涎腺 (颌下腺、腮腺 )组织作对照。采用免疫组化方法 ,检测组织中TIP30蛋白的表达。用显色指数和灰度值两个指标表示TIP30蛋白的表达量。结果 TIP30蛋白在正常涎腺组织中高表达 ,显色指数为强阳性 ;在2 0例肿瘤组织及淋巴结转移组织中 ,TIP30蛋白呈低表达 ,80 % (1 6 / 2 0 )为阴性 ;1 6例显色指数 <5 % ,4例为弱阳性。TIP30在肿瘤组织中表达的灰度值为 1 86 4 5 ,在正常涎腺组织中表达的灰度值为 1 2 3 2 5 ,两者相比有显著性差异 (P <0 0 1 )。 结论 抑癌基因TIP30蛋白在SACC组织中表达量与正常涎腺组织相比显著性降低 ,由此可推论TIP30基因在SACC中表达是缺失的
Objective To detect the expression of tumor suppressor gene TIP30 in Salivary Adenoid Cystic Carcinoma (SACC). Methods Totally 20 salivary adenoid cystic carcinoma specimens from 1983 to the present in our hospital were selected. There were 9 males and 11 females, and the other 8 normal salivary glands (submandibular gland and parotid gland) were selected as controls. The expression of TIP30 protein was detected by immunohistochemistry. The expression level of TIP30 protein was expressed by two indexes of color rendering index and gray value. Results TIP30 protein was overexpressed in normal salivary gland tissues, and the color index was strongly positive. TIP30 protein was lowly expressed in 20 cases of tumor tissues and lymph node metastasis, and 80% (16/20) of them were negative. Cases of color index <5%, 4 cases were weakly positive. The gray value of TIP30 expression in tumor tissues was 1 86 45, and the expression of TIP30 in normal salivary glands was 1 2 3 2 5, which showed significant difference (P <0.01). Conclusion The expression level of TIP30 protein in SACC tissues is significantly lower than that in normal salivary gland tissues. It can be deduced that the expression of TIP30 gene in SACC is absent
