为探讨脑损伤诱导神经细胞凋亡的作用及其机制,应用大鼠液压冲击脑损伤模型,在脑损伤后不同时间段(3h、12h、24h、48h、72h)处死动物,分别应用HE染色、流式细胞仪检测、TUNEL法测定、DNA凝胶电泳分析、免疫组化技术研究神经细胞凋亡的改变。结果显示:动物经中度脑损伤处理后,其受伤各时间段脑组织可见凋亡的神经细胞及TUNEL阳性细胞, DNA呈现“梯状”断裂现象,细胞凋亡百分率为9.8%~14.0%(对照组17%),凋亡相关基因(cmyc,fas,fasL)表达明显增加。以上研究提示,脑外伤能够诱导神经细胞凋亡,其作用机制可能与其诱导某些凋亡相关基因的表达有关。
To investigate the effect and mechanism of brain injury-induced apoptosis in neurons, rats were impacted by fluid percussion brain injury model and sacrificed at different time points (3h, 12h, 24h, 48h, 72h) Flow cytometry, TUNEL assay, DNA gel electrophoresis analysis, immunohistochemistry to study the change of neuronal apoptosis. The results showed that apoptotic neurons and TUNEL positive cells were observed in the brain tissue of rats after moderate brain injury. The DNA showed “ladder-like” fracture with the percentage of apoptotic cells ranging from 9.8% to 14.0% Control group 17%), the expression of apoptosis-related genes (cmyc, fas, fasL) increased significantly. These studies suggest that traumatic brain injury can induce neuronal apoptosis, the mechanism may be related to its expression of some apoptosis-related genes.
