目的观察神经前体细胞表达的发育性下调基因9(NEDD9)在食管癌中的表达情况,干扰EC109食管癌细胞NEDD9后观察食管癌细胞侵袭、迁移能力的改变,明确NEDD9在人食管癌细胞系中的功能。方法通过免疫组织化学实验检测食管癌组织及其癌旁组织中NEDD9的表达;通过RNA干涉(RNAi)技术干扰NEDD9后,运用反转录PCR、Western blot技术检测NEDD9的干扰效率;下调NEDD9表达后,MTT法检测食管癌细胞的增殖,TranswellTM实验检测EC109细胞侵袭、迁移能力;Western blot法检测细胞增殖细胞核抗原(PCNA)、Bax和Bcl-2蛋白水平。结果食管癌组织中NEDD9阳性表达率明显高于癌旁组织。NEDD9-siRNA转染12、24 h后,转染组增殖抑制率、侵袭率和迁移率均明显降低。Bcl-2表达降低,Bax表达升高。结论 NEDD9在食管癌中表达显著高于癌旁组织,NEDD9能够影响癌细胞的侵袭迁移能力。
Objective To observe the expression of NEDD9 in esophageal cancer and to investigate the effect of NEDD9 on esophageal cancer cell line NEDD9 after esophageal cancer cell invasion and migration. In the function. Methods Immunohistochemistry was used to detect the expression of NEDD9 in esophageal squamous cell carcinoma tissues and its adjacent tissues. After interfering with NEDD9 by RNA interference (RNAi) technology, the interference efficiency of NEDD9 was detected by reverse transcription PCR and Western blot. The expression of NEDD9 was down-regulated The proliferation of esophageal cancer cells was detected by MTT assay. The invasion and migration of EC109 cells were detected by TranswellTM assay. The protein expressions of proliferating cell nuclear antigen (PCNA), Bax and Bcl-2 were detected by Western blot. Results The positive rate of NEDD9 in esophageal cancer tissues was significantly higher than that in paracancerous tissues. After transfected with NEDD9-siRNA for 12,24 h, the proliferation inhibition rate, invasion rate and migration rate of transfection group were significantly decreased. Bcl-2 expression decreased, Bax expression increased. Conclusion NEDD9 expression in esophageal cancer was significantly higher than that in paracancerous tissues, and NEDD9 could affect the invasion and migration of cancer cells.
