为了解决五四○六菌种传代繁殖容易发生退化的问题,我们探索用冰冻法进行复壮的可能性。在冰冻环境下使强的孢子选择出来,弱孢子则死亡淘汰,使菌种得以复壮。试验表明在-15℃和-30℃低温下,冰冻处理五四○六抗生菌孢子悬液和琼脂斜面菌种,能促进菌种生长繁殖。冰冻处理的菌种经移植8代后,生长优势尚未有明显的减退。-30℃处理的菌种生长快,培养2天菌苔已铺满琼脂斜面,培养3天,菌苔局部呈现粉红色。培养5天后,菌苔全部呈现粉红色,菌苔丰满,有露珠分泌,冰片味较浓。
In order to solve the problem of degeneration likely to occur in 5460 bacteria passage and reproduction, we explore the possibility of rejuvenation with the method of freezing. In the frozen environment so that strong spores are selected, the death of weak spores are eliminated, so that strains can be rejuvenated. Experiments show that at -15 ℃ and -30 ℃ low temperature, freeze treatment of 5460 antibacterial spores suspension and agar slant bacteria can promote the growth and reproduction of bacteria. Frozen strains after 8 generations of transplants, the growth advantage has not yet significantly reduced. The strain cultured at -30 ℃ grew rapidly. After 2 days of cultivation, the lawn was covered with agar slant and cultured for 3 days. After 5 days of cultivation, all of the lawn moss appeared pink, the lawn was full, there was dewdrop secretion, and the borneol smell was thicker.
