目的:研究利用雌激素诱导人胆管上皮细胞的增殖作用,并探讨其临床作用。方法:用17β-雌二醇作用于人胆管上皮细胞,CCK8方法检测17β-雌二醇对人胆管上皮细胞生长的影响,Westernblot胆管上皮细胞的雌激素受体(ER-α,ER-β)表达水平,并通过ELISA检测上述两种培养条件下胆管上皮细胞培养上清中表皮细胞生长因子(epidermal growth factor,EGF)的分泌水平。结果:17β-雌二醇可以促进人胆管上皮细胞增殖,雌二醇组的人胆管上皮细胞ER-α,ER-β表达水平明显高于对照组(P<0.05),用ELISA检测两组的上清中表皮细胞生长因子(epidermal growth factor,EGF)无显著性差异(P>0.05)。结论:17β-雌二醇可以诱导人胆管上皮细胞增殖,主要通过胆管上皮细胞的雌激素受体表达水平上升,而不是表皮细胞生长因子的通路来实现的。
Objective: To study the effect of estrogen on the proliferation of human bile duct epithelial cells and to explore its clinical role. Methods: The effects of 17β-estradiol on the growth of human bile duct epithelial cells were detected by CCK8 assay. The expressions of estrogen receptor (ER-α, ER-β) in Westernbiliary bile duct epithelial cells The level of epidermal growth factor (EGF) secretion in bile duct epithelial cell culture supernatant was detected by ELISA. Results: 17β-estradiol promoted the proliferation of human bile duct epithelial cells. The expression of ER-α and ER-β in human bile duct epithelial cells in estradiol group was significantly higher than that in control group (P <0.05) There was no significant difference in epidermal growth factor (EGF) in the supernatant (P> 0.05). CONCLUSION: 17β-estradiol can induce the proliferation of human bile duct epithelial cells mainly through the increase of estrogen receptor expression in biliary epithelial cells, but not the epidermal growth factor pathway.
