目的观察依达拉奉(edaravone,ED)联合川芎嗪(tetramethylpyrazine,TMP)对脓毒血症大鼠肝线粒体超氧化物歧化酶(superoxide dismutase,SOD)和羟自由基(hydroxyl,·OH)的影响。方法 Sprague-Dawley(SD)大鼠30只分为假手术组、手术对照组、联合治疗组(下称治疗组),手术对照组和治疗组均予盲肠结扎穿刺法(caecurm ligated and punctured,CLP)制作大鼠脓毒血症模型;假手术组只轻轻挤压盲肠,不结扎穿刺;3组均于术前15 min及术后3h均皮下注射乳酸左氧氟沙星20mg/kg。治疗组于术前15min及术后3h皮下注射ED 5mg/kg和TMP 15mg/kg。3组术后18h采取肝组织测SOD、·OH,并对肝组织进行显微镜观察。结果手术对照组大鼠肝组织显微镜下可见肝细胞浊肿,水样变,假手术组和治疗组大鼠肝组织病理改变不明显。手术对照组肝线粒体·OH活力与假手术组比较明显升高(P<0.01),SOD活力与假手术组比较明显降低(P<0.05);治疗组肝线粒体·OH活力与手术对照组比较明显降低(P<0.05),SOD活力与手术对照组比较明显升高(P<0.01),差异均有统计学意义。结论 ED联合TMP能降低脓毒血症大鼠肝线粒体·OH活力,升高肝线粒体SOD活力,提高清除自由基和抗氧化能力,具有保护肝功能作用,值得临床推广使用。
Objective To observe the effects of edaravone (ED) combined with tetramethylpyrazine (TMP) on superoxide dismutase (SOD) and hydroxyl radical (OH) in liver mitochondria of septic rats, influences. Methods Thirty Sprague-Dawley (SD) rats were randomly divided into sham operation group, operation control group, combination therapy group (hereinafter referred to as treatment group), operation control group and treatment group by caecurm ligated and punctured ). The sepsis model was made in rats. The sham operation group only gently pressed the cecum and did not ligate the puncture. All the three groups were subcutaneously injected with levofloxacin 20 mg / kg 15 min before and 3 h after operation. The treatment group received subcutaneous injection of ED 5 mg / kg and TMP 15 mg / kg 15 min before operation and 3 h after operation. Twenty-eight hours after operation, SOD and .OH in liver tissue were measured and the liver tissues were observed under microscope. Results The hepatocytes were found in the liver of the operation control group under microscope. The pathological changes of the liver tissue were not obvious in the sham operation group and the treated group. The activity of mitochondrial · OH in the liver of the operation group was significantly higher than that of the sham-operated group (P <0.01), while the activity of SOD was significantly lower than that of the sham operation group (P <0.05) (P <0.05). The SOD activity was significantly increased compared with the control group (P <0.01), and the differences were statistically significant. Conclusions ED combined with TMP can reduce · · OH activity of hepatic mitochondria of septic rats, increase SOD activity of liver mitochondria, improve the ability of scavenging free radicals and anti-oxidation, protect liver function, and is worthy of clinical promotion.
