为了解胎肝提取液中造血生长因子对急性髓细胞白血病(AML)细胞的作用,将胎肝提取液经超滤后分成中分子蛋白(MMWP)和低分子蛋白(LMWP),利用体外集落培养法比较两者的活性,实验结果表明:当MMWP在1250、250、50和10倍稀释时对K_(562)细胞株和AML祖细胞具有明显的刺激增殖作用,作用优于LMWP.进一步比较MMWP与rHuGM-CSF两者作用活性,结果在无PHA-LCM存在时,两者均需较高的浓度才能使CFU-AML生长良好。表明白血病祖细胞对GM-CSF的敏感性低于正常细胞,而存在PHA-LCM时,较低剂量的MMWP和常规剂量的rHuGM-CSF(200u/ml)就能显著地提高CFU-AML集落产率。提示MMWP具有类似于rHuGM-CSF的作用,通过对CFU-AML的刺激增殖作用,提高其对化疗药物的敏感性。
