目的:探讨氟伐他汀对人外周血单个核细胞(MNC)中抗炎蛋白膜联蛋白A1表达水平的影响,为氟伐他汀抗炎抗动脉粥样硬化机制提供新的视点。方法:在体外分离培养人MNC,分别用不同浓度氟伐他汀(0.2、0.4、0.8mg/L)诱导不同时间(2、8、16、24h)后,应用流式细胞仪检测MNC膜联蛋白A1表达水平。结果:氟伐他汀可使MNC膜联蛋白A1表达上调(P<0.01),呈浓度、时间依赖方式。阴性对照组平均荧光强度(MFI)2.4000,0.2mg/L组MFI3.2950,0.4mg/L组4.0225,0.8mg/L组4.8000;2h组4.2625,8h组4.2100,16h组4.0900,24h组4.0225。结论:氟伐他汀可以通过上调MNC膜联蛋白A1表达发挥抗炎、抗动脉粥样硬化作用。
Objective: To investigate the effect of fluvastatin on the expression of anti-inflammatory protein Annexin A1 in human peripheral blood mononuclear cells (MNC), and to provide a new perspective for the anti-inflammatory anti-atherosclerotic mechanism of fluvastatin. Methods: Human MNC was isolated and cultured in vitro. Fluvastatin (0.2, 0.4, 0.8 mg / L) was used to induce MNC annexin A1 expression level. Results: Fluvastatin increased MNC Annexin A1 expression (P <0.01) in a concentration-dependent and time-dependent manner. The average fluorescence intensity (MFI) of negative control group was 2.4000, the MFI of 2.2950 of 0.2mg / L group, 4.0225 of 0.4mg / L group and 4.8000 of 0.8mg / L group were 4.2625 for 2h group, 4.2100 for 2h group, 4.02100 for 16h group, 4.0900 for 16h group, 4.0225 for 24h group. Conclusion: Fluvastatin can exert anti-inflammatory and anti-atherosclerotic effects by up-regulating MNC Annexin A1 expression.
