目的探讨原发性肝细胞肝癌(HCC)患者肝癌组织中核因子-κB抑制物激酶ε(IKKε)、磷酸化核因子-κB抑制物激酶ε(p-IKKε)的表达与患者临床病理特征及生存期的关系。方法收集2006年12月至2008年8月64例手术切除的HCC组织标本,采用免疫组化染色技术检测肝癌及癌旁组织中IKKε、p-IKKε蛋白的表达。对IKKε、p-IKKε的表达水平与临床分期、转移之间的关系进行单因素分析。采用Kaplan-Meier分析法对其进行生存评估。对患者进行随访1~160周(平均64.90周)。结果 IKKε与p-IKKε蛋白在癌旁组织中表达均不明显,但在肝癌组织中均高表达。IKKε的阳性表达与肿瘤分化程度(P=0.049)、临床分期(P=0.039)显著相关,而与年龄、性别、血清甲胎蛋白(AFP)、肿瘤大小以及有无转移无相关性(P均>0.05)。p-IKKε的阳性表达与肿瘤分化程度(P=0.017)、有无转移(P=0.031)显著相关,而与年龄、性别、血清AFP、肿瘤大小以及临床分期无相关性(P均>0.05)。与IKKε表达阴性患者比较,IKKε表达阳性的HCC患者其生存时间显著降低(P=0.035)。结论IKKε、p-IKKε蛋白在肝癌组织中的表达明显高于癌旁组织;且IKKε阳性表达的HCC患者生存时间明显缩短,pIKKε阳性表达与HCC患者生存时间无明显相关性。IKKε的检测可作为HCC的预后判断指标。
Objective To investigate the expression of nuclear factor-κB inhibitor ε (IKKε) and phosphorylated nuclear factor-κB inhibitor ε (p-IKKε) in primary hepatocellular carcinoma (HCC) patients and its clinicopathological characteristics and survival Period of the relationship. Methods 64 cases of HCC tissue samples from December 2006 to August 2008 were collected for immunohistochemical staining for the expression of IKKε and p-IKKε in hepatocellular carcinoma and its adjacent tissues. Univariate analysis was performed on the relationship between the expression level of IKKε, p-IKKε and clinical stage and metastasis. Survival was assessed using Kaplan-Meier analysis. Patients were followed up for 1 to 160 weeks (mean, 64.90 weeks). Results The expression of IKKε and p-IKKε protein in adjacent non-cancerous tissues was not significant, but both were highly expressed in HCC tissues. The positive expression of IKKε was correlated with tumor differentiation (P = 0.049) and clinical stage (P = 0.039), but not with age, sex, serum AFP, tumor size and metastasis > 0.05). The positive expression of p-IKKε was significantly correlated with tumor differentiation (P = 0.017) and metastasis (P = 0.031), but not with age, gender, serum AFP, tumor size and clinical stage . Compared with patients with negative expression of IKKε, the survival time of patients with IKKε-positive HCC was significantly lower (P = 0.035). Conclusions The expression of IKKε and p-IKKε protein in HCC tissues is significantly higher than that in paracancerous tissues. The survival time of IKKεpositive HCC patients was significantly shortened. The positive expression of pIKKε had no significant correlation with the survival time of HCC patients. IKKε detection of HCC can be used as a prognostic indicator.
