目的:观察糖皮质激素肾阴虚证模型各靶组织相关激素受体的表达变化以及滋肾阴方的干预作用。方法:实验分为4组,正常组、模型组、模型+滋肾阴复方组,正常+滋肾阴复方组。模型组、模型+滋肾阴复方组采用氢化可的松腹腔注射50mg/kg,连续7天,造成大鼠肾阴虚证模型。采用Western blot检测各组大鼠下丘脑、胸腺、性腺的糖皮质激素受体α(GRα)、甲状腺激素受体α/β(TRα/β)、雄激素受体(AR)的表达。结果:糖皮质激素造模导致大鼠体重降低、肾上腺、胸腺重量降低;也致GRα、TRα/β、AR的表达改变,使GRα表达在下丘脑下调,在胸腺上调。滋肾阴方使用后,能显著保护动物体重、胸腺重量,肾上腺重量有明显上升趋势;并能显著降低GR在胸腺的表达,升高AR在性腺的表达,升高TRα/β在胸腺的表达。结论:糖皮质激素肾阴虚模型相应靶组织受体表达也发生了显著改变,在研究补肾中药时,需要注意研究其在受体和受体后水平的效应,而不仅仅关注对血浆激素水平的影响。
Objective: To observe the changes of the expression of related hormone receptors in each target tissue of glucocorticoid-induced kidney yin deficiency model and the intervention of Zishenyin prescription. Methods: The experiment was divided into 4 groups, normal group, model group, model + Zishen Yin Prescription group, normal + Zishenyin Prescription group. Model group, model + Zishenyin Compound group were given intraperitoneal injection of hydrocortisone 50mg / kg, for 7 days, resulting in rat kidney yin deficiency model. Western blot was used to detect the expressions of GRα, TRα / β and AR in the hypothalamus, thymus and gonads of rats in each group. Results: Glucocorticoid induced weight loss, decreased weight of adrenal gland and thymus, and altered the expression of GRα, TRα / β and AR. GRα expression was down-regulated in the hypothalamus and up-regulated in the thymus. Zishenyin prescription can significantly protect animal body weight, thymus weight and adrenal weight, and can significantly reduce the expression of GR in the thymus, increase the expression of AR in the gonads, and increase the expression of TRα / β in the thymus . CONCLUSION: The expression of the corresponding target tissue receptor in glucocorticoid-induced deficiency of kidney-yin model also changed significantly. When studying kidney-tonifying traditional Chinese medicine, attention should be paid to the effect on the level of receptor and receptor, not only on the level of plasma hormone Impact.
