镍化合物可在DNA中诱导生成各种小的氧化产物。本研究,利用~(32)P后标记法测定镍(Ⅱ)化合物是否可在体内外诱导大的DNA加合物,离体研究中,DNA与NiCl_2和H_2O_2在37℃下培养1h,可检测到2种主要加合物和几种次要加合物。在培养0~24h和NiCl_2浓度为0~800μM范围内时,这2种主要加合物可随时间和浓度增加而增加,加合物产量在经去羟自由
Nickel compounds can induce various small oxidation products in DNA. In this study, we used ~ (32) P post-labeling method to determine whether nickel (II) compounds could induce large DNA adducts in vivo or in vitro. DNA, NiCl 2 and H 2 O 2 were cultured at 37 ℃ for 1 h, To 2 main adducts and several minor adducts. When cultured for 0-24 h and NiCl 2 concentration ranged from 0-800 μM, the two main adducts could increase with time and concentration, adduct yield increased after dehydrogenation
