利用来自巴西橡胶树(Hevea brasiliensis Muell.Arg)未成熟花药的愈伤组织进行了体细胞胚胎发生的研究。采用附加BAP、NAA及2,4-D各1mg/L的改良MS培养基诱导获得带有明显前胚结构的胚性愈伤组织。将这些愈伤组织再转移到含有1mg/L激动素及0.1mg/LIAA的培养基上进行培养继而获得体细胞胚胎。胚性愈伤组织的组织学研究结果显示,体细胞胚胎产生于愈伤组织外表的分生组织细胞。此外,在愈伤组织诱导及体细胞胚胎发生中均观察到克隆变异的发生。
Somatic embryogenesis was studied using callus from immature anthers of Hevea brasiliensis Muell. Arg. The embryogenic callus with obvious pre-embryo structure was induced by modified MS medium supplemented with 1mg / L BAP, NAA and 2,4-D. These callus tissues were further transferred to a medium containing 1 mg / L kinetin and 0.1 mg / L IAA to obtain a somatic embryo. Histological studies of embryogenic callus revealed that somatic embryos are produced from meristematic cells that are external to the callus. In addition, clonal variation was observed in callus induction and somatic embryogenesis.
